The genetic structure of 128 Japanese-flounder(Paralichths olivaceus Temminck et Schlegel)of 3 cultured groups in Rongcheng and Yantai of Shandong province, Beidaihe of Hebei province in China was analyzed using PCR-RFLP of the mitochondrial D-loop region。 We have found the high variation in mtDN A D-Loop of Japanese flounder: 1 the variation of the D-loop region length: in terms of the size of fragments, we grouped the samples into 5 groups, they are Ⅰ (2。 4kbp)\Ⅱ(2。 5kbp)\Ⅲ(2。 5kbp)\Ⅳ(2。 6kbp)and Ⅴ(2。 8kbp)。 The Kb values of Rongcheng, Beidaihe and Yantai were 0。 5178, 0。 7888, 0。 8006, respectively; 2At the same time, we have firstly found the heteroplasmic phenomenon in Japanese flounder (Paralichthys olivaceus Temminck et Schlegel)。 Among the Beidaihe groups, there were 12 two-fragments-contained specimen (29。 27% of the groups); 6 three-fragments-contained specimen (16。 43% of the groups), who is the most heteroplasmic contained groups; 3 The variation of the haplotype: Through the analysis of the restriction fragments, we gained 29 haplotypes and 11 polymorphic sites of 29 restriction sites。 From the 29 haplotypes, there were 20 haplotypes only occurred in one fish, namely the characteristic haplotype of this speciman。 The haplotype diversity of Rongcheng 0。8152 was the lowest, Beidaihe 0。 9289 was the highest。 The average Shannon's indices of Rongcheng, Beidaihe and Yantai were 1。 4832,2。 4240 and 2。 4066, respectively。 It is the first Domestic report on the genetic structure research of Japanese flounder that using this Technology。
