百两金皂苷A和B微生物转化研究

朱砂根为紫金牛科紫金牛属植物朱砂根(Ardisia crenata Sims)的根。性味苦、辛、凉,具有清热解毒、散瘀止痛的作用。百两金皂苷A和B(ardisiacrispinA和ardisiacrispin B)是从中药朱砂根中分离得到的主要三萜皂苷类活性成分之一。利用微生物的方法筛选24种真菌菌株对百两金皂苷A和B的生物转化,其中有五种菌株对其有转化作用,文中选定两种进行放大累积分离,得到转化产物8个,通过化学和光谱学方法鉴定了其中7个转化产物的结构。它们分别是:西克拉明皂苷元A-3β-O-{α-D-半乳吡喃糖基-(1→4)-[β-D-木吡喃糖基-(1→2)]-β-D-葡萄吡喃糖基-(1→4)-[β-D-葡萄吡喃糖基-(1→2)]-α-L-阿拉伯吡喃糖苷) Cyclamiretin A-3β-O-{α-D-galactopyranosyl-(1→4)-[β-D-xylopyranosyl-(1→2)]-β-D-glucopy- ranosyl-(1→4)-[α-D-glucopyranosyl-(1→2)]-α-L-arabinopyranoside)(2)*,西克拉明皂苷元 A-3β-O-{α-D-半乳吡喃糖基-(1→4)-β-D-葡萄吡喃糖基-(1→2)-[β-D-木吡喃糖基-(1→2)- β-D-葡萄吡喃糖基-(1→4)]-α-L-阿拉伯吡喃糖苷}Cyclamiretin A-3β-O-{α-D-galactopy- ranosyl-(1→4)-β-D-glucopyranosyl-(1→2)-[β-D-xylopyranosyl-(1→2)-β-D-glucopyranosyl- (1→4)]-α-L-arabinopyranoside}(3)*,西克拉明皂苷元A-3β-O-{α-D-半乳吡喃糖基-(1→4)-[α-L-鼠李吡喃糖基-(1→2)]-β-D-葡萄吡喃糖基-(1→4)-[β-D-葡萄吡喃糖基-(1→2)]-α-L-阿拉伯吡喃糖苷}Cyclamiretin A-3β-O-{α-D-galactopyranosyl-(1→4)-[α-L- rhamnopyranosyl-(1→2)]-β-D-glucopyranosyl-(1→4)-[β-D-glucopyranosyl-(1→2)]-α-L-arabin- opyranoside)(4)*,西克拉明皂苷元A-3p-O-{α-L-鼠李吡喃糖基-(1→2)-β-D-葡萄吡喃糖基-(1→4)-[β-D-葡萄吡喃糖基-(1→2)]-[α-D-半乳吡喃糖基-(1→3)]-α-L-阿拉伯吡喃糖苷> Cyclamiredn A-3β-O-{α-L-rhanmopyranosyl-(1→2)-β-D-glucopyranosyl-(1→4)-[β-D-gluco- pyranosyl-(1→2)]-[α-D-galactopyranosyl-(1→3)]-α-L-arabinopyranoside}(5) * ,西克拉明皂苷元A-3β-O-{β-D-木吡喃糖基-(1→2)-β-D-葡萄吡喃糖基-(1→4)-[β-D-葡萄吡喃糖基 -(1→2)]-[α-D-半乳吡喃糖基-(1→3)]-α-L-阿拉伯吡喃糖苷}Cyclamiretin A-3β-O- {β-D-xylopyranosyl-(1→2)-β-D-glucopyranosyl-(1→4)-[β-D-glucopyranosyl-(1→2)]-[α-D- galactopyranosyl-(1→3)]-α-L-arabinopyranoside)(6)*,西克拉明皂苷元A-3β-O-[β-D-木吡喃糖基-(1→2)-β-D-葡萄吡喃糖基-(1→4)-α-L-阿拉伯吡喃糖苷]Cyclamiretin A-3β-O-[β-D- xylopyranosyl-(1→2)-β-D-glucopyranosyl-(1→4)-α-L-arabinopyranoside](7),西克拉明皂苷元A-3β-O-[α-L-鼠李吡喃糖基-(1→2)-β-D-葡萄吡喃糖基-(1→4)-α-L-阿拉伯吡喃糖苷] Cyclamirctin A-3β-O-[α-L-rhamnopyranosyl-(1→2)-β-D-glucopyranosyl-(1→4)-α-L-arabino- pyranoside)](8)。除转化产物7和8外,其余均为未见文献报道的新转化产物。其中对百两金皂苷A和B有转化能力的两种菌株中,菌株燕麦曲霉(Aspergillus avenaceus,AA3.4454)对百两金皂苷A和B有选择性的使其糖苷链上易增加一个α-D-半乳糖,菌株长柄链格孢(Alternaria longipes,AL 3.2875)能够使百两金皂苷A和B水解掉末端的葡萄糖成为连有三个糖的降糖产物。将转化产物2、3、4、5、7、8用MTT法对MCF-7(人乳腺癌细胞)和Hela(宫颈癌细胞)两种肿瘤细胞进行体外细胞毒活性测试,均有一定的诱导肿瘤细胞凋亡的活性。
