    Electrospun nanofibers based on carboxymethyl cellulose/polyvinyl alcohol as a potential antimicrobial wound dressing

    查看更多>>摘要:? 2022In this work, citric acid-based quantum dots (CA-QDs) as a novel and safe crosslinked agent was applied in different feeding ratios (5–15 wt%) to synthesize carboxymethyl cellulose/polyvinyl alcohol (CMC/PVA) nanofibers (NFs) for the first time. Colistin (CL) as an antibacterial agent was also loaded (2 w/w%) during the synthesizing process of CMC/PVA electrospun NFs to trigger antimicrobial properties. The morphological, hydrophilic, and mechanical properties of the prepared NFs were fully investigated with different techniques. The electrospun NFs with crosslinking ratios of 10 wt% CA-QDs revealed appropriate mechanical properties. According to cell culture data, the prepared NFs demonstrated good cytocompatibility against HFF-1 cells (over 80% cell viability). Remarkably, CL-loaded NFs showed desired antibacterial efficacy against S. aureus, E. coli, K. pneumoniae, and P. aeruginosa with 1.0–1.4, 1.3–1.4, 0.8–1.0, and 1.3–1.5 cm inhibition zones, respectively. These outcomes suggested that the fabricated NFs can be useful as wound healing scaffolds.

    Tunable mechanical behavior of collagen-based films: A comparison of celluloses in different geometries

    查看更多>>摘要:? 2022Collagen (Col) films were reinforced by celluloses in different geometries: microcrystalline cellulose (MCC), cellulosic fines (CF), cellulose nanofiber (CNF) and cellulose nanocrystals (CNC). The reinforcement mechanisms were investigated by the elastoplasticity and fracture appearance. Compared with the fracture stress of collagen film (67.5 MPa), the Col-CNF films effectively borne the stress (95.8 MPa) by intercrystalline fracture, ascribing the abundant hydrogen bonding and mechanical locking between cellulose and collagen. The toughness of Col-CF films was increased by the interfibrillar slippage of CF and pull-off of CF within the matrix, improving the strain-to-break from 8.37% to 12.13%. The films added with MCC and CNC weaken the mechanical behavior, due to the defects and lack of mechanical locking. Besides, the effects of celluloses' geometries on the thickness, density, water-tightness, thermal stability, crystallinity and FTIR of films were also investigated. These provide the evidence that the geometries of fillers diversely improve the behaviors of collagen film offering strategies for the film with adjustable mechanical properties.

    Effect of the structural features of biobased linear polyester plasticizers on the crystallization of polylactides

    查看更多>>摘要:? 2022 The AuthorsThis work presents, for the first time, a detailed report on how the nucleation and crystallization of polylactide (PLLA) are affected by biobased aliphatic polyesters plasticizers. Three biobased polyesters were synthesized via solvent-free two-stage melt polycondensation of adipic acid (AdA) with three different biobased aliphatic diols and used as plasticizers for poly (L-lactic acid) (PLLA). The molecular structure of the synthesized polyesters was proved using 1H NMR, 13C NMR and Fourier transform infrared (FTIR) spectroscopy. PLLA/AdA-based blends containing 10 wt% of the polyester plasticizers were studied by tensile tests, dynamic mechanical analysis (DMA), wide-angle x-ray scattering (WAXS), differential scanning calorimetry (DSC) and polarized light optical microscopy (PLOM). Adding the plasticizers to PLLA decreased Tg by up to 11 °C and significantly increased the elongation at break by about 8 times compared with neat PLLA. The addition of 10 wt% of any AdA-based plasticizer to PLLA increases the nucleation rate from the glassy state by around 50–110 % depending on the plasticizer. The overall crystallization rate from the glassy state was 2–3 times faster for the plasticized PLLAs than neat PLLA. These results are a consequence of the lower energy barrier for both nucleation and growth processes. The incorporation of AdA-based linear polyesters had an incremental impact on the crystal growth rate (or secondary nucleation) of PLLA spherulites from the melt and glassy states. In conclusion, the AdA-based aliphatic polyesters allowed to enhance PLLA crystallization rates and showed interesting potential for the formulation of fully biobased PLLA blends.

    Dopamine receptor (DAR) and dopa decarboxylase (DDC) mediate hepatopancreas antibacterial innate immune reactions in Procambarus clarkii

    查看更多>>摘要:? 2022 Elsevier B.V.Dopa is decarboxylated by dopa decarboxylase (DDC) to form dopamine, which is a significant signaling molecule in the neuroendocrine system. The dopamine receptor (DAR) is an important transmembrane receptor responsible for receiving extracellular signals in the DAR-mediated signaling pathway. In the present study, the expression patterns of Pc-dar were investigated after bacterial challenge. The obviously changed expression patterns showed Pc-dar was related to the antibacterial innate immunity. Endogenous Pc-DDC enzymatic activities were obviously downregulated after Pc-ddc dsRNA injection. The expression level of Pc-dar mRNA was obviously upregulated after bacterial injection when the expression level of Pc-ddc was knocked down. In addition, the upregulation trend of endogenous Pc-DDC enzymatic activities was obviously restrained after bacterial stimulation when Pc-ddc was knocked down. Finally, melanization was downregulated in crayfish hemolymph compared with the dsGFP injection group. In the RNAi assay, the results of qRT-PCR showed that Toll (TLRs) signaling pathway-related genes were activated in the early stages of bacterial stimulation when Pc-ddc was knocked down. Four tested ROS-related antioxidant enzyme genes were significantly upregulated after bacterial challenge compared with the dsGFP injection group. The above results indicated that Pc-DDC and Pc-DAR play important mediating roles in the neuroendocrine immune (NEI) system of crayfish.

    Structural features of biologically active extracellular polysaccharide produced by green microalgae Dictyosphaerium chlorelloides

    查看更多>>摘要:? 2022 Elsevier B.V.Ion-exchange chromatography of the biologically active extracellular biopolymer produced by D. chlorelloides yielded ten fractions differing in yield, protein content, monosaccharide composition and molecular weight distribution. Their sugar compositional analyses showed rhamnogalactans, substituted to different extent by mannose and glucose, as a dominant EPS component in all fractions (91 %) except one containing arabinogalactan (7 %). In highly branched rhamnogalactans the quantity of linear (1,3-; 1,4- and 1,6-linked) and branched β-D-galactose units (1,3,6-, 1,4,6- and 1,3,4,6-linked) was nearly equal. From various α-L-rhamnose linkages the 1,2,4-linkage was dominant. Data indicate a rhamnogalactan backbone of EPS, branched by terminal mannose and glucose units, and a lot of O-methylated derivatives of galactose residues (2-O-methyl, 2,3-O-dimethyl, 3-O-methyl and 6-O-methyl). In individual fractions their content and type varied. Detail study of the arabinogalactan showed that its backbone consists of 1,3-linked β-D-Galp units; some of them are branched through O-4 by 6-OMe-α-D-Galp- (1 → 2) -α-L-Araf side chain, other through O-6 by 3-OMe-β-D-Galp, 6-OMe-β-D-Galp, β-D-Galp and β-D-Galf.

    PBP1 plays key roles in sex pheromone reception of the fall armyworm

    查看更多>>摘要:? 2022Spodoptera frugiperda among the China population employs a four-component sex pheromone blend to accelerate male-female allocation and mating behavior. The underlying molecular mechanism has been incompletely elucidated. In the current study, we showed that differences existed between genders toward the four sex pheromone components, including Z9–14:AC, Z7–12:AC, Z9–12:AC, and Z11–16:AC, in terms of electrophysiological responses and behavioral valences. Male adults were significantly more sensitive to all tested compounds than female adults. Furthermore, ecological outputs may be related to four pheromone-binding proteins, namely, SfruPBP1, SfruPBP2, SfruPBP3, and SfruPBP4. They formed four distinct clades within the lepidopteran phylogeny, and male adults expressed significantly higher levels of SfruPBP1 and SfruPBP2 than female adults. We observed the highest binding affinities of SfruPBP1 toward all four sex pheromone components. SfruPBP4 had moderate binding affinities for Z7–12:AC, Z11–16:AC, and Z9–12:AC, while SfruPBP2 showed binding toward Z9–14:AC. This observation suggests that SfruPBP1 plays a key role in sex pheromone discrimination and drives sexually biased behavioral decisions toward certain pheromone components. These findings will help to develop behavioral-mediating tools as part of integrated pest management approaches for this cross-border pest.

    Fabrication of uniform lignin nanoparticles with tunable size for potential wound healing application

    查看更多>>摘要:? 2022 Elsevier B.V.The construction of lignin nanoparticles (LNPs) with both lignin properties and nanomaterial properties through controlling the morphologies and structures of lignin is one of the effective ways to realize its application in the field of biomedicine. Firstly, the morphology and chemical structure of LNPs were studied in detailed. The results showed that the chemical structural characteristics of LNPs had not changed significantly and its morphology was more regular shape and narrower size distribution (50–350 nm). Besides, LNPs also exhibited excellent water dispersion stability and high negative zeta potential. Subsequently, LNPs as wound dressings had good antioxidant property, excellent adsorption capacity of protein, outstanding bactericidal activity and remarkable biocompatibility, suggesting that LNPs did not interfere with cell proliferation during wound healing. Finally, the in vivo results of mouse wounds further illustrated that treatment of wounded skin wounds with LNPs enhanced its effective healing. After 15 days, as compared with the untreated control and original lignin (OL) groups, the wounds treated of LNPs was completely closed and granulation tissue formation was advanced. Overall, this study can be a good method for high-value applications of LNPs, and highlighting the advantages of using lignin as medical adjuvant nanomaterials to accelerate wound healing.

    Curcumin-loaded HKUST-1@ carboxymethyl starch-based composites with moisture-responsive release properties and synergistic antibacterial effect for perishable fruits

    查看更多>>摘要:? 2022The spoilage of fruit is one of the most important causes of fruit waste. High humidity by fresh fruit respiration leads to bacterial reproduction, which is the key factor of products corruption. Herein, a biological multifunctional film (Cur-HKUST-1@CMS/PVA) for fruits preservation with a high moisture environment was developed by cross-linking carboxymethyl starch (CMS)/polyvinyl alcohol (PVA) with MOF-199 (HKUST-1), and loaded with curcumin. The hydrophilic CMS facilitates water adsorption and moisture can stimulate curcumin release from HKUST-1. HKUST-1 not only acts as curcumin carriers but also forms synergistic antibacterial with curcumin to improve the antibacterial activity of the composites. XRD and SEM demonstrated that moisture disrupts the structure of HKUST-1 and releases curcumin and the results showed that the release of curcumin increased from 25.11 % to 58.32 % after moisture stimulation. In addition, Cur-HKUST-1@CMS/PVA had excellent antibacterial activity and antioxidant ability. As validation, the film can keep pitaya and avocado freshness at least 4 days longer than the control, confirming the effectiveness of Cur-HKUST-1@CMS/PVA in preventing fruit decay. Consequently, Cur-HKUST-1@CMS/PVA is a promising active packaging material for improve the shelf life of perishable fruits.

    Peculiarities of the interaction of sodium dodecyl sulfate with chitosan in acidic and alkaline media

    查看更多>>摘要:? 2022In this work, the interaction between the negatively charged surfactant sodium dodecyl sulfate (SDS) and partially N-reacetylated chitosan (RA-CHI), which is soluble at pH range up to pH 12, is studied in a wide pH range including alkaline media by light scattering (LS) and isothermic titration calorimetry (ITC). It is shown that in the weakly alkaline medium (pH 7.4), RA-CHI/SDS interaction is exothermic and cooperative. This interaction is found to be coupled with proton transfer from the buffer substance to chitosan as it is revealed by the dependence of the measured heat release on the ionization enthalpy of the buffer. At higher pH values (pH > 8), another mechanism of interaction is observed that include SDS micellization induced by hydrophobic interactions with polymer segments, so that no phase separation occurred in these mixtures. The results obtained can contribute to expand the knowledge about application of chitosan for preparation of pharmaceutical and cosmetic compositions containing anionic surfactants.

    Complete secretion of recombinant Bacillus subtilis levansucrase in Pichia pastoris for production of high molecular weight levan

    查看更多>>摘要:? 2022 Elsevier B.V.Three signal peptides from α-mating factor (α-MF), inulinase (INU) and native levansucrase (LS) were compared for secretion efficiency of Bacillus subtilis levansucrase SacB-T305A in Pichia pastoris GS115. The first complete secretion of bacterial levansucrase in yeasts under methanol induction was achieved while using α-MF signal. The secreted recombinant Lev(α-MF) proved to be glycosylated by combination of NanoLC-MS/MS and Endo H digestion. Interestingly, glycosylation not only improved significantly the polymerase thermostability, but also reversed the products profiles to favor synthesis of high molecular weight (HMW) levan which accounted for approximately 73 % to total levan-type polysaccharides. It indicated for the first time that the glycosylation of recombinant B. subtilis levansucrase affected significantly the products molecular weight distribution. It also provided a promising enzymatic way to effectively product HMW levan from sucrose resources.
