    Large-scale survey for canine vector-borne parasites in free-ranging dogs and foxes from six diverse bioclimatic regions of Chile

    查看更多>>摘要:Chile is a large country with a marked range of climate conditions that make it an ideal scenario for the study of vector-borne parasites (VBPs); however, knowledge about their distribution is limited to a few confined areas of this country. The presence of Hepatozoon spp., piroplasmids, Leishmania spp. and filarioids was investigated through molecular and serological methods in blood and serum samples of 764 free-ranging rural dogs, 154 Andean foxes (Lycalopex culpaeus), and 91 South American grey foxes (Lycalopex griseus) from six bioclimatic regions across Chile. Hepatozoon spp. DNA was exclusively detected in foxes (43% prevalence), including se-quences closely related to Hepatozoon felis (24.1%; only Andean foxes), Hepatozoon americanum (16.2%; only grey foxes), and Hepatozoon canis (1.25%; in one grey fox). Risk factor assessment identified a higher probability of Hepatozoon infection in juvenile foxes. DNA of piroplasmids was detected in 0.7% of dogs (Babesia vogeli) but in no fox, whilst antibodies against Babesia sp. were detected in 24% of the dogs and 25% of the foxes, suggesting a wider circulation of canine piroplasmids than previously believed. A positive association between the presence of antibodies against Babesia and high Rhipicephalus sanguineus sensu lato burden was observed in dogs. Leish-mania spp. DNA and antibodies were detected in 0.8% and 4.4% of the dogs, respectively. Acanthocheilonema reconditum was the only blood nematode detected (1.5% of the dogs and no fox). Differences in prevalence among bioregions were observed for some of the VBPs. These results expand our knowledge about the occurrence of vector-borne parasites in Chile, some of which are firstly reported herein. This information will facilitate the diagnosis of vector-borne diseases in domestic dogs and improve the control measures for both domestic and wild canids.

    查看更多>>摘要:Wild birds have an important role as hosts of ticks infected by rickettsiae. However, the role of birds as reservoirs of tick-borne rickettsiae is unknown and poorly understood. This is particularly relevant in several tropical and subtropical areas, where migration influences the global spread of ectoparasites and pathogens of public health importance. This research aimed to detect and evaluate the exposure to spotted fever group rickettsiae in wild birds that could represent reservoirs in the Department of Arauca in the Colombian Orinoquia region. Sampling was conducted in three municipalities of the Department of Arauca (Colombia). Blood samples were collected from 255 birds and processed to obtain serum (n = 155) and DNA (n = 255) samples. The serum samples were processed for indirect immunofluorescence assays (IFA) for the detection of antibodies to Rickettsia rickettsii, Rickettsia parkeri, Rickettsia amblyommatis, Rickettsia rhipicephali, and Rickettsia bellii. Additionally, we investigated rickettsiae DNA in blood samples by amplification of the citrate synthase gene (gltA). The IFA results revealed seropositivity in 97 samples from 54 species of resident and migratory birds. No sample was positive for rickettsial DNA. The presence of antibodies in 62.5% of the sera indicates previous exposure of these birds to rickettsiae. The null detection of rickettsiae in the blood of seropositive birds is possibly due to a short period of bacteremia. Experimental studies are required to improve our understanding of the role of wild birds as sources of rickettsial infections in ticks.

    查看更多>>摘要:Dogs are important in the epidemiology of tick-borne diseases, playing a significant role mainly in endemic areas of rickettsiosis and borreliosis, and serving as sentinels in epidemiological surveys. We analyzed the distribution of Rickettsia and Borrelia spp. in dogs and their ticks in the municipalities of Guaxupe'& nbsp;, Minas Gerais, and Tapiratiba, Sao Paulo, Brazil, two areas non-endemic for Baggio-Yoshinari Syndrome (BYS) and rickettsiosis. Serum from 242 dogs of different ages (> 12 months) and breeds were tested by the Indirect Immunofluorescence Assay (IFA) using specific antigens for Rickettsia rickettsii, Rickettsia parkeri, Rickettsia amblyommatis, Rickettsia rhipicephali, and Rickettsia bellii, and by an indirect Enzyme Linked Immunosorbent Assay (ELISA) for the detection of homologous IgG antibodies against Borrelia burgdorferi American strain G39/40. Ticks were collected from the animals and subjected to PCR and nested PCR for detection of Rickettsia spp. (synthase citrate gene) and Borrelia spp. (flagellin gene). Serological data showed that 7.85% (19/242) of the analyzed dogs were seropositive for at least one of the five Rickettsia antigens tested; one (0.41%) was considered seropositive for R. parkeri (1:64) and one (0.41%) for R. rickettsii (1:256). Nine (3.72%) were considered seropositive for R. bellii and one (0.41%) for R. amblyommatis; a seroprevalence of 13.64% (33/242) for IgG class immunoreactive antibodies against B. burgdorferi was observed. A total of 148 ticks were collected from the dogs; among these 3.40% were identified as larvae of the genus Rhipicephalus and 0.70% of the genus Amblyomma; 89.8% were identified as Rhipicephalus sanguineus, 4.70% Amblyomma sculptum and 1.40% Amblyomma ovale; all negative for PCR of Rickettsia spp. and nested PCR for Borrelia spp.. The serological findings of this study suggest the circulation of Rickettsiae associated with the spotted fever group and vector ticks, just like Borrelia spp. in a non-endemic Brazilian area, drawing attention to the possibility of a zoonotic cycle in the region.

    Farmers' perception on the control of gastrointestinal parasites in organic and conventional sheep production in Sweden

    查看更多>>摘要:A questionnaire was sent to sheep owners in Sweden to get information about anthelmintic drug use. The survey also investigated how respondents experienced problems with gastrointestinal nematode infections (GIN) focusing on Haemonchus contortus. The response rate was 31% and included both conventional and organic farms. The use of anthelmintics was low (45%), among which a majority (76%) drenched ewes on a single occasion, mostly with ivermectin (59%) followed by albendazole (19%). Other drugs were used rarely, however, unawareness of GIN risk was high (19%), especially among respondents with few animals. Anthelmintic dose calculations were done after visual appraisal by 63% and 22% calibrated the equipment before drug delivery, which is worrying since underdosing is a risk factor for the development of anthelmintic resistance. Like with anthelmintics, the perceived risk for GIN increased with herd size both by conventional and organic farmers. Faecal examination for the presence of GIN was done by 65% of the respondents and, among their sheep, H. contortus was or had been diagnosed in 41% of the herds. Irrespective of new stock had been imported from other countries or not, common problems were reported by 5% and 7% of the organic and conventional producers, respectively. Land use and grazing management strategies differed more in relation to herd size than by production form, with a majority (47%) having their sheep grazed in several paddocks, or at least the lambs were moved when separated from the ewes at weaning (25%). In contrast set stocked grazing was mainly reported on smaller farms. Co-grazing with cattle and horses were also frequently reported irrespective of production form, but with cattle to a somewhat greater degree on larger organic farms. Wild cervids, especially roe deer, were frequently observed on sheep pastures (87%). The veterinary involvement was higher on organic (65%) than on conventional farms (53%), and only 5% considered advice unimportant. Still, some conventional and organic producers treated sheeps routinely without a prior diagnosis, against the national regulations. 46% of the respondents drenched new and replacement stock. In conclusion, although some differences were observed between conventional and organic producers, the divergences were mainly due to herd size categories. Furthermore, despite a high veterinary involvement, we identified factors which can contribute to anthelmintic use, such as poor quarantine procedures, and deworming routines that can contribute to anthelmintic resistance in H. contortus.

    查看更多>>摘要:Co-infection by two paramphistome species, Orthocoelium sp. and Paramphistomum epiclitum, is found in cattle in Thailand. The morphological features of these and other paramphistomes under a light microscope are similar, resulting in misidentification and misdiagnosis. We classified these paramphistomes into three morphological variation types, namely Orthocoelium sp., P. epiclitum MV1 (immature), and P. epiclitum MV2 (matured). Ten morphological characteristics were investigated, and the values were transformed into 25 ratio criteria for statistical investigation. Morphometric analysis can classify the variation of these specimens using differences in the bifurcal level, the vitellaria starting level, the starting level of the anterior testis, and the center level of the posterior testis positions by body length ratios. These ratios can separate the samples into three morphologically different groups, whereas molecular analysis based on the nuclear internal transcribed spacer 2 (ITS2) region and the mitochondrial cytochrome c oxidase subunit I (COI) gene could only distinguish two specific groups. In addition, the Orthocoelium specimen, related to O. dicranocoelium and O. parvipapillatum according to morphological and histological analysis, was monophyletic grouped via ITS2 analysis. Our study provides a scientific basis for the taxonomic classification and clustering of morphologically varying species, improving the identification, detection, and diagnosis of co-infecting paramphistomes.

    Iatrogenic doramectin overdosing causes toxicity in sheep: A case report

    查看更多>>摘要:Macrocyclic lactones are widely used endectocides in ruminants, with a high margin of safety for labeled indications. No previous report of iatrogenic doramectin overdosing has been published. We report an outbreak in a sheep flock in Northeast Patagonia, Argentina. Toxicity signs were observed in more than 10% and 59% of ewes and lambs, respectively, particularly those with low body condition, treated with doramectin 3.5% long-acting injectable formulation, presumably at the indicated dose of 700 mu g/kg. Clinical signs included lethargy, mydriasis and coma. Doramectin concentration in blood samples was 826.8 (& PLUSMN;119.3) ng/ml. Cerebrospinal fluid (CSF) and liver doramectin concentration in euthanized lambs were 3.26-4.28 ng/ml and 8506-8772 ng/g, respectively. Epidemiological and clinical information, and high doramectin concentration were sufficient to confirm the neurotoxicity. Scarce fat deposition could have altered doramectin pharmacokinetic which may have accounted for the observed neurotoxicity. Special care should be taken when animals under similar nutritional conditions are treated with macrocyclic lactones.
