    Effect of different modeling time on intestinal bacteria in letrozole induced PCOS rats

    查看更多>>摘要:Objective: The differences of ovarian morphology, reproductive hormones, glucose and lipid metabolism and intestinal bacteria in rats with polycystic ovary syndrome (PCOS) induced by triazole were compared. Method: Eighteen 21 SPF female SD rats were randomly divided into group A (3-week group), group B (5-week group) and group D (control group) by random number table.Group A received letrozole + CMC-Na mixture by gavage in the first 3 weeks and CMC-Na solution by gavage in the last 2 weeks, group B received letrozole + CMC-Na mixture by gavage for 5 weeks, and group D received CMC-Na solution by gavage for 5 weeks, and all three groups of rats were fed with normal diet.At the end of gavage, the body weight of rats in each group was observed, the histological changes of ovaries were observed by hematoxylin-eosin (HE) staining, the serum levels of estradiol (E2), follicle stimulating hormone (FSH), luteinizing hormone (LH), testosterone (T), total cholesterol (TC), triglyceride (TG), fasting blood glucose (Glu), fasting insulin (FINS) and lipopolysaccharide (LPS) were measured by enzyme-linked immunosorbent assay (ELISA), and the LH/FSH ratio and insulin resistance index (HOMA IR) were calculated; the intestinal bacteria of rats were detected by 16S rRNA technique. Result: 1. Comparison of ovary histomorphology: Under light microscope, multiple luteum and oocytes were observed in mature follicles in group D, and granulosa cells were orderly arranged and multilayered, without cystic dilated follicles. There were no mature follicles in the ovarian tissues of group A and GROUP B. The follicles were irregular in structure and more cystic dilated follicles were visible. The number of granular cells in some follicles decreased or even disappeared. 2. Comparison of sex hormone levels: compared with group D, T level in group B was significantly increased (P < 0.001), and T level in group A had an upward trend (P > 0.05); The LH/FSH levels in group A and B were significantly increased (P < 0.001; P < 0.001). Compared with group A, E2 in group B was significantly decreased (P < 0.05) and T was significantly increased (P < 0.01). 3. Comparison of glucose and lipid metabolism levels: Compared with group D, TC levels in groups A and B were significantly increased (P < 0.01; P < 0.01). Compared with group A, TG in group B was significantly increased (P < 0.05). There were no significant differences in Glu, FINS and HOMA-IR levels among all groups. 4. Comparison of LPS levels: Compared with group D, the serum LPS levels of rats in groups A and B were significantly increased (P < 0.001; P < 0.01). 5. Intestinal flora analysis and comparison: At the phylum level, compared with group D, the abundance of Firmicutes in group B increased (P < 0.01), Firmicutes in group A showed an upward trend (P > 0.05), and the abundance of Bacteroidetes in groups A and B decreased (P < 0.05). At the genus level, compared with group D, Lactobacillus in group B increased (P < 0.01). The results of LEfSe analysis showed that there were differences in the composition of various intestinal bacteria among the three groups (LDA > 3).Conclusion: The phenotype of PCOS rats was related to the length of modeling, and the phenotypic characteristics of PCOS in rats at 5 weeks of modeling were more typical than those in rats at 3 weeks of modeling; PCOS can cause changes in intestinal flora, and the changes in the structure of intestinal flora between groups are related to different modeling duration.

    查看更多>>摘要:Objective: To investigate the effect of Fufei Gushen Decoction on airway inflammation and glucocorticoid receptor in rats with chronic obstructive pulmonary disease. Methods: Fifty Wistar male rats were randomly divided into 5 groups: blank control group, COPD model group, Fufei Gushen Yin high, medium and low dose groups, 10 rats in each group, except the blank control group, the remaining 4 groups were Smoked combined with lipopolysaccharide (LPS), cold air stimulation to create CODP rat model. After successful modeling, the blank control group and COPD model group were fed with distilled water 3ml/only, Fufei Gushen Yin high, medium and low dose groups were given 1.02, 0.51, 0.26g Chinese medicine granules/100g/day, respectively. 2 times a day for 28 consecutive days. Samples were collected, hematoxylin-eosin (HE) staining was used to observe the pathological changes of lung tissue, and enzyme-linked immunosorbent assay (ELISA) was used to detect the tumor necrosis factor alpha (TNF-α) in the serum and right alveolar lavage fluid (BALF) of rats in each group. ), the content of transforming growth factor β1 (TGF-β1), interleukin-17 (IL-17A) and matrix metalloproteinase (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in the left lung tissue The expression level of real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) rat left lung tissue GRmRNA, immunohistochemistry (IHC) to determine the expression level of left lung tissue glucocorticoid receptor (GR). Results: The content of TNF-α, TGF-β1 and IL-17A in the serum of COPD rats in Fufei Gushen Yin high, medium and low dose groups and BALF were significantly reduced compared with the COPD model group (P<0.05); The expressions of TIMP-1 and MMP-9 in tissues were lower than those in COPD model group (P<0.05); the expressions of GRmRNA and GR in lung tissues were higher than those in COPD model group (P<0.05), and were higher in Fufei Gushen Yin Among the middle- and low-dose groups, the middle-dose group has the most significant effect. Conclusion: Fufei Gushen Decoction can inhibit the release of inflammatory factors in lung tissue of COPD rats, improve airway inflammation and remodeling, and increase hormone sensitivity.

    查看更多>>摘要:Objective: To prepare the liposomes of mangrove oil, Optimization of the formulation of mangrove oil liposomes by Box Behnken response surface methodology.Methods: Preparation of Rhododendron oil liposomes by ethanol injection probe ultrasound, Determination of gemacrone by HPLC. The ratio of lecithin to cholesterol (X1), drug lipid ratio (X2) and phospholipid concentration (X3) were used as independent variables, and encapsulation efficiency (Y) was used as dependent variable, the formulation was optimized by Box Behnken response surface method, and the entrapment efficiency was predicted. The entrapment efficiency, particle size, polydispersity index (PDI), Zeta potential and drug loading of the optimized liposomes were evaluated. Results: The optimal prescription and preparation of Folium Rhododendri Daurici oil liposome was confirmed as follows: X1=7.28:1、X2=11.34:1、X3=9.32mg·mL-1,the encapsulation efficiency was(82.55±1.66)%, the particle size was (130.531 ± 46) nm, the polydispersity index was 0. 185 ± 05, Zeta potential was (21. 970 ± 36) mV, the drug loading was (5.941±0.12)%. Conclusion: The Box Behnken response surface method is accurate to obtain the optimal formulation of mangrove oil liposomes, it has high precision and good prediction effect. And the preparation process of mangrove oil liposomes is stable and feasible.

    Effect of TRAF6 gene silencing on hepatocellular carcinoma cell line SMCC7721 and its possible mechanism

    查看更多>>摘要:Objective: To explore the effect of TRAF6 gene silencing on the function of hepatocellular carcinoma SMCC7721 and its possible mechanism. Method: Cell lines were constructed by cell transfection technology and verified by quantitative real-time PCR. Cell functional changes were observed by CCK8 method, Transwell test and Method of EdU. Western blotting was used to explore the possible mechanism of action. Result: TRAF6 RNA was abnormally up-regulated in HCC, and TRAF6 levels were detected in both HCC cell lines and L02 cells. SMCC7721 was selected as TRAF6 high expression cell. The results of CCK8 assay and EdU method showed that the decrease of TRAF6 expression significantly inhibited the proliferation of SMCC7721 cells. The results of CCK8 assay and EdU method showed that the decrease of TRAF6 expression significantly inhibited the proliferation of SMCC7721 cells. Overexpression of TRAF6 in TRAF6 knockdown cells can restore and enhance cell proliferation. Transwell assay confirmed that the invasiveness of SMCC 7721 cells treated with siRNA was significantly reduced. After treatment with LV-Rescue plasmid, the cell invasion was restored and enhanced. Western blotting showed that the protein levels of YB-1, Wnt, β-catenin, c-myc and Cyclin D1 were significantly down-regulated in siRNA group. On the contrary, the expression level of CYLD protein increased. Conclusion: As an important intracellular junctive protein in tumor cells, TRAF6 may improve the expression of pro-cancer factors C-myc and Cyclin D1 by modifying (ubiquitination) YB-1, thus improving the proliferation ability of cells. This process may be closely positively correlated with the Wnt/β-catenin pathway, and negatively correlated with the expression of CYLD protein.

    SNAPC3 gene rs12093 polymorphism is significantly associated with ischemic stroke wind syndrome

    查看更多>>摘要:Objective: To investigate whether The small nuclear RNA activating complex polypeptide 3 (SNAPC3) gene rs12093 polymorphism is associated with ischemic stroke (IS) and Traditional Chinese Medicine (TCM) syndromes of IS. Methods: This study enrolled 774 patients with ischemic stroke (IS) and 793 normal controls. Massarray technology was used for genotyping. And genetic association analysis was estimated by PLINK program. Results: ①SNAPC3 gene rs12093 polymorphism was significantly associated with the risk of ischemic stroke wind syndrome of IS [OR=0.76, 95%CI(0.58-0.99), P=0.047 in dominant model)]. ②After adjusting age and gender, SNAPC3 gene rs12093 polymorphism and the score of ischemic stroke wind syndrome(Padj=0.037 in additive model;Padj=0.009 in recessive model), the correlation is statistically significant. ③After adjusting age and gender, platelet (PLT) (Padj=0.024 in addiction model; Padj=0.039 in dominant model)and thrombin time(TT)(Padj=0.042 in dominant model) were significantly associated with rs12093 polymorphism in patients with ischemic stroke wind syndrome. Conclusion: SNAPC3 gene rs12093 polymorphism may affect the occurrence of ischemic stroke wind syndrome.

    Relationship between platelet-lymphocyte ratio and carotid atherosclerosis in middle-aged and elderly patients with type 2 diabetes mellitus

    查看更多>>摘要:Objective: To investigate the relationship between platelet-lymphocyte ratio (PLR) and carotid atherosclerosis (CAS) in middle-aged and elderly patients with T2DM. Methods: A total of 420 middle-aged and elderly patients with T2DM who were hospitalized in the Endocrinology Department of our hospital from September 2018 to September 2020 and were examined by ultrasound were selected as the subjects. The carotid artery intima-media thickness (CIMT) was examined by color Doppler ultrasonography, including normal CIMT group (n=111), CIMT thickening group (n=103) and CAS plaque group (n=206). The differences of serum PLR levels among the three groups were compared. Multivariate Logistic regression was used to analyze the relationship between PLR and CAS plaque formation. The receiver operating curve (ROC) was used to evaluate the value of multivariate Logistic regression model (Logit P) in the diagnosis of CAS lesions. Results: The mean value of PLR in CAS plaque group (127.86±48.54) was significantly higher than that in CIMT normal group (109.15±48.35). Multivariate Logistic regression analysis showed that after adjusting for other confounding factors, the increase of PLR was an independent risk factor for the occurrence of CAS plaque. ROC showed that the efficacy of multivariate Logit P model in the diagnosis of CAS plaque was (AUC=0.807,95%CI=0.759-0.858). Conclusions: PLR is one of the important risk factors for the occurrence of CAS lesions. At the same time, the multivariate Logistic regression model established in this study has clinical reference value in predicting CAS lesions in middle-aged and elderly patients with T2DM.

    查看更多>>摘要:Objective: To study the potential therapeutic effect of Yigan Powder on Alzheimer's disease (AD) comorbid Depressive Disorder with Network Pharmacology. Methods: The active ingredients of Yigan Powder were screened from Traditional Chinese Medicine Systems Pharmacology database and analysis platform (TCMSP) by ADME parameters, targets are predicted by Swiss Target Prediction database, disease targets are downloaded from GeneCards, OMIM and PharGKB database. Using R to run GO terms enrichment and KEGG pathway enrichment analysis. Protein interaction data is downloaded from the String database. Cytoscape software is used to build network. AutoDock Vina and PyMOL software are used for molecular docking. Results: There are 125 active ingredients in Yigan Powder with 953 predicted targets, 85 of predicted targets are related to Alzheimer's Disease comorbid Depressive Disorder, shows highly enriched signaling pathways and biological processes. PPI network shows APP, MAPK1 and STAT3 may be important potential treatment of Alzheimer's Disease comorbid Depressive Disorder. The results of AutoDock Vina docking showed that the active ingredients of Yigan Powder had good binding activity with important receptors. Conclusion: Yigan Powder shows effect on neurotransmitter metabolism, synaptic transmission, neuroinflammation and other aspects in the treatment of Alzheimer's Disease comorbid Depressive Disorder.

    Rules of acupuncture point selection in the treatment of rheumatoid arthritis through data mining

    查看更多>>摘要:Objective: To explore the rules of acupuncture point selection in thetreatment of rheumatoid arthritis by applying data mining and modern Statistical software.Methods: We retrieved literature from China hownet(CNKI), VIP database(VIP), Wanfang,PubMed, MEDLINE, Embase database from the founding of the database to February 1,2020. The main information of meridians and acupoints were entered into Excel, SPSS22.0 was used to extract data and do clustering analysis. Results: Quchi, Zusanli, Hegu, Yanglingquan, Xiyan were often chosen for electropuncture treatment of rheumatoid arthritis. Most of the acupoints are located in the large intestine channel of hand yangming bladder meridian or bladder channel of foot Taiyang, Shaoyang gallbladder channel of foot, stomachmeridian of foot Yangming, the energizer meridian and concentrated in the upper and lower extremities. The frequency of use of five Shu points, crossing point, source point, lowerconfluent and eight meeting points acupoints is higher. Cluster analysis can divide the included acupoints into 5 clusters. Conclusion: Quchi, Zusanli, Hegu, Yanglingquan, Xiyan are often chosen for electropuncture treatment of rheumatoid arthritis. Most of the selected acupoints aremostly distributed in the acupoints which are located in the large intestine channel of hand yangming bladder meridian or bladder channel of foot Taiyang, Shaoyang gallbladder channel of foot, Stomach meridian of foot Yangming, and the energizer meridian.

    Treatment of diabetic kidney disease of stage Ⅲ-Ⅳ with Yiqi Wenyang Huoxue therapy: A meta-analysis

    查看更多>>摘要:Objective: To systematically evaluate the effect of Yiqi Wenyang Huoxue therapy in treating diabetic kidney disease(DKD) of stage Ⅲ-Ⅳ. Method: The relevant literature was retrieved by computer from CNKI, Wanfang and VIP databases,CBM,PubMed,EMBase,Cochrane Library and Web of Science. Articles screening and data extraction were performed by two researchers separately and independently. The Meta-analysis was conducted with RevMan 5.3. Results: A total of 10 randomized controlled trials were collected,including 778 cases with DKD. Meta-analysis showed that the Yiqi Wenyang Huoxue Method combined with western medicine basic treatment could reduce 24hUTP[MD=-0.36, 95%CI (-0.46, -0.26), P <0.00001], UAER[MD=-36.72,95%CI (-47.21, -26.23), P<0.00001], Scr[MD= -16.28, 95%CI(-20.59,-11.97),P<0.00001],BUN[MD=-1.45,95%CI(-1.76,-1.14), P <0.00001], TC[MD=-0.64, 95%CI(-1.07,-0.21), P=0.004].There was also a reduction in HbA1c[MD=-0.50,95%CI (-0.94, -0.05),P =0.03], but the difference is not significant. Conclusion: Based on the current evidence, for the treatment of DKD of stage Ⅲ-IV, basic western medicine treatment combined with Yiqi Wenyang Huoxue method may better reduce urine protein, total cholesterol levels and improve renal function. The conclusion of this study needs to be further researched by high-quality studies, because that the quality of the included studies is generally low.

    Clinical experience of Professor Xie Jingri in the deficiency-cold of spleen and stomach type of complex peptic ulcer

    查看更多>>摘要:Objective: To summarize Professor Xie's clinical experience in treating the deficiency cold of spleen and stomach type of complex peptic ulcer. Methods: through the observation of the curative effect of the patients with the deficiency cold of spleen and stomach type of complex peptic ulcer in the outpatient department of liver, spleen and stomach department of our hospital, the dialectical application and clinical experience of Professor Xie Jingri's self-made prescription in clinical practice were summarized. Results: Professor Xie Jingri's self-made prescription can effectively improve the clinical symptoms of patients with the deficiency cold of spleen and stomach type of complex peptic ulcer, and through individual syndrome differentiation and treatment, make the self-made prescription vary from person to person, clinical addition and subtraction, so as to further improve the clinical efficacy of patients. Conclusion: Professor Xie Jingri's self-made prescription is effective in treating complex peptic ulcer of spleen stomach deficiency cold type, which is worthy of further clinical application and promotion.
