    A human-specific insertion promotes cell proliferation and migration by enhancing TBC1D8B expression

    查看更多>>摘要:Human-specific insertions play important roles in human phenotypes and diseases.Here we reported a 446-bp insertion(Insert-446)in intron 11 of the TBC1D8B gene,located on chromosome X,and traced its origin to a portion of intron 6 of the EBF1 gene on chromosome 5.Interestingly,Insert-446 was present in the human Neanderthal and Denisovans genomes,and was fixed in humans after human-chim-panzee divergence.We have demonstrated that Insert-446 acts as an enhancer through binding transcript factors that promotes a higher expression of human TBC1D8B gene as compared with orthologs in macaques.In addition,over-expression TBC1D8B promoted cell pro-liferation and migration through"a dual finger"catalytic mechanism(Arg538 and Gln573)in the TBC domain in vitro and knockdown of TBC1D8B attenuated tumorigenesis in vivo.Knockout of Insert-446 prevented cell proliferation and migration in cancer and normal cells.Our results reveal that the human-specific Insert-446 promotes cell proliferation and migration by upregulating the expression of TBC1D8B gene.These findings provide a significant insight into the effects of human-specific insertions on evolution.

    Nitrogen deposition mediates more stochastic processes in structuring plant community than soil microbial community in the Eurasian steppe

    查看更多>>摘要:Anthropogenic environmental changes may affect community assembly through mediating both deterministic(e.g.,competitive exclusion and environmental filtering)and stochastic processes(e.g.,birth/death and dispersal/colonization).It is traditionally thought that en-vironmental changes have a larger mediation effect on stochastic processes in structuring soil microbial community than aboveground plant community;however,this hypothesis remains largely untested.Here we report an unexpected pattern that nitrogen(N)deposition has a larger mediation effect on stochastic processes in structuring plant community than soil microbial community(those<2 mm in diameter,including archaea,bacteria,fungi,and protists)in the Eurasian steppe.We performed a ten-year nitrogen deposition experiment in a semiarid grassland ecosystem in Inner Mongolia,manipulating nine rates(0-50 g N m-2 per year)at two frequencies(nitrogen added twice or 12 times per year)under two grassland management strategies(fencing or mowing).We separated the compositional variation of plant and soil microbial communities caused by each treatment into the deterministic and stochastic components with a recently-developed method.As nitrogen addition rate increased,the relative importance of stochastic component of plant community first increased and then decreased,while that of soil microbial community first decreased and then increased.On the whole,the relative importance of stochastic component was significantly larger in plant community(0.552±0.035;mean±standard error)than in microbial community(0.427±0.035).Consistently,the proportion of compositional variation explained by the deterministic soil and community indices was smaller for plant community(0.172-0.186)than microbial community(0.240-0.767).Meanwhile,as nitrogen addition rate increased,the linkage between plant and microbial community composition first became weaker and then became stronger.The larger stochasticity in plant community relative to microbial community assembly suggested that more stochastic strategies(e.g.,seeds addition)should be adopted to maintain above-than below-ground biodiversity under the pressure of nitrogen deposition.

    Degeneration of foundation cushion species induced by ecological constraints can cause massive changes in alpine plant communities

    查看更多>>摘要:Foundational cushion plants can re-organize community structures and sustain a prominent proportion of alpine biodiversity,but they are sensitive to climate change.The loss of cushion species can have broad consequences for associated biota.The potential plant community changes with the population dynamics of cushion plants remain,however,unclear.Using eight plant communities along a climatic and community successional gradient,we assessed cushion population dynamics,the underlying ecological constraints and hence associated plant community changes in alpine communities dominated by the foundational cushion plant Arenaria polytrichoides.The population dynamics of Arenaria are attributed to ecological constraints at a series of life history stages.Reproductive functions are constrained by increasing associated beneficiary plants;subsequent seedling establishment is constrained by temperature,water and light availability,extreme climate events,and interspecific competition;strong competitive exclusion may accelerate mortality and degeneration of cushion populations.Along with cushion dynamics,species composition,abundance and community structure gradually change.Once cushion plants completely degenerate,previously cushion-dominated communities shift to relatively stable communities that are overwhelmingly dominated by sedges.Climate warming may accelerate the degeneration process of A.polytrichoides.Degeneration of this foundational cushion plant will possibly induce massive changes in alpine plant communities and hence ecosystem functions in alpine ecosystems.The assessment of the population dynamics of foundation species is critical for an effective conservation of alpine biodiversity.

    Genomic data and ecological niche modeling reveal an unusually slow rate of molecular evolution in the Cretaceous Eupteleaceae

    查看更多>>摘要:Living fossils are evidence of long-term sustained ecological success.However,whether living fossils have little molecular changes remains poorly known,particularly in plants.Here,we have introduced a novel method that integrates phylogenomic,comparative genomic,and ecological niche modeling analyses to investigate the rate of molecular evolution of Eupteleaceae,a Cretaceous relict angiosperm family endemic to East Asia.We assembled a high-quality chromosome-level nuclear genome,and the chloroplast and mitochondrial genomes of a member of Eupteleaceae(Euptelea pleiosperma).Our results show that Eupteleaceae is most basal in Ranunculales,the earliest-diverging order in eudicots,and shares an ancient whole-genome duplication event with the other Ranunculales.We document that Eupteleaceae has the slowest rate of molecular changes in the observed angiosperms.The unusually low rate of molecular evolution of Eupteleaceae across all three independent inherited genomes and genes within each of the three genomes is in association with its conserved genome architecture,ancestral woody habit,and conserved niche requirements.Our findings reveal the evolution and adaptation of living fossil plants through large-scale environmental change and also provide new insights into early eudicot diversification.

    Global multifaceted biodiversity patterns,centers,and conser-vation needs in angiosperms

    查看更多>>摘要:The Convention on Biological Diversity seeks to conserve at least 30%of global land and water areas by 2030,which is a challenge but also an opportunity to better preserve biodiversity,including flowering plants(angiosperms).Herein,we compiled a large database on dis-tributions of over 300,000 angiosperm species and the key functional traits of 67,024 species.Using this database,we constructed biodi-versity-environment models to predict global patterns of taxonomic,phylogenetic,and functional diversity in terrestrial angiosperms and provide a comprehensive mapping of the three diversity facets.We further evaluated the current protection status of the biodiversity centers of these diversity facets.Our results showed that geographical patterns of the three facets of plant diversity exhibited substantial spatial mismatches and nonoverlapping conservation priorities.Idiosyncratic centers of functional diversity,particularly of herbaceous species,were primarily distributed in temperate regions and under weaker protection compared with other biodiversity centers of taxonomic and phylogenetic facets.Our global assessment of multifaceted biodiversity patterns and centers highlights the insufficiency and unbalanced conservation among the three diversity facets and the two growth forms(woody vs.herbaceous),thus providing directions for guiding the future conservation of global plant diversity.
