    Construction of a HERG mutant L539fs/47-*558W pEGFP vector and the expression of the fusion protein in HEK293 cells

    查看更多>>摘要：Objective:To construct a human ether-a-go-go-related gene(HERG)nonsense mutant L539fs/47-*558W into the autonomously fluorescent,eukaryotic expression vector pEGFP-C2,and to verify expression of the reconstruct in human embryonic kidney-293(HEK293)cells.Methods:The mutational fragment was subcloned into pEGFP-C2-HERG by double digestion of Sbf Ⅰ,Eco91 Ⅰ and rejoining of T4 ligase.After verification,the recombinant pEGFP-C2-L539fs/47-*558W and pEGFP-C2-HERG were respectively transfected into HEK293 cells for 48 h by the Lipofect method to observe the expression location of the fusion protein by laser confocal imaging scanning in vivo.pcDNA3-L539fs/47-*558W and pcDNA3-HERG were transfected to observe the expression location of the HERG protein by immunofluoresceoce.The mutant protein size was determined by Western blotting.Results:The about 1 kb-sized mutation region cDNA fragment from pcDNA3-L539fs/47-* 558W and the about 7.2 kb-sized target vector fragment from pcDNA3-HERG were ligated after purification and gel recovery.pEGFP-C2-L539fs/47*-558W,approximately 8.2 kb,was demonstrated successfully been constructed under agarose gel electrophoresis and further sequencing.Laser confocal imaging showed that pEGFP-C2-HERG was mainly expressed in the membrane,whereas truncated mutant-type HERG in the pEGFP-C2 vector was partially located in the cytoplasm,the others were transported to the cell membrane in living HEK293 cells.The same as the immunofluoresceoce results after transfection of pcDNA3-HERG and pcDNA3-L539fs/47-558W.Wild-type HERG-GFP fusion protein expressed 160 and 180 kDa bands.The mutant and mutant-GFP fusion proteins were 70 and 100 kDa,respectively.Conclusion:pEGFP-C2-L539fs/47-*558W was successfully constructed by double digestion method GFP had no effect on its protein expression and trafficking in HEK293 cells,which laid a foundation for the further study on L539fs/47-*558W.

    Risk factors for post-hepatectomy liver dysfunction in primary liver cancer patients with concurrent hepatic schistosomiasis and chronic hepatitis

    查看更多>>摘要：Objective:The purpose of this study was to analyze risk factors for development of post-hepatectomy liver dysfunction in primary liver cancer(PLC)patients with concurrent hepatic schistosomiasis and chronic hepatitis.Methods:A retrospective analysis of 73 PLC patients with concurrent hepatic schistosomiasis and chronic hepatitis,of which 16 patients developed liver dysfunction(persistent ascites or pleural effusion or occurrence of liver-related potentially fatal complications)following hepatectomy,was performed.After clinical characteristics were recorded,preoperative liver function parameters and surgery-related parameters in these patients were assessed.Seventeen potential risk factors for post-hepatectomy liver dysfunction were identified.The association between these potential risk factors and post-hepatectomy liver dysfunction then was analyzed.Results:Univariate analysis showed that liver cirrhosis,intraoperative blood loss,and preoperative total bilirubin were associated with the development of post-hepatectomy liver dysfunction.Multivariate logistic regression analysis of these three factors revealed that intraoperative blood loss ≥600 mL and cirrhosis were two independent risk factors for post-hepatectomy liver dysfunction in PLC patients with concurrent hepatic schistosomiasis and chronic hepatitis.Conclusion:Keeping intraoperative blood loss below 600 mL can help avoid the development of post-hepatectomy liver dysfunction in liver cancer patients with concurrent hepatic schistosomiasis and chronic hepatitis.For patients with concomitant liver cirrhosis,every effort should be made to minimize potential liver function impairment induced by other adverse factors.

    Living alone,health and preventive care use among the elderly in Shanghai,China

    查看更多>>摘要：Objective:The purpose of the study was to compare the preventive care use and health between the elderly living alone and living with other individuals and identify strategies to improve the preventive care use among the elderly living alone.Methods:A questionnaire including socioeconomic characteristics,preventive care use,health status and Social Support Rate Scale was administered to 240 elderly living alone and 244 elderly living with other individuals in Shanghai,China.Logistic regression analysis was used to examine the predictors of preventive care use.Results:The elderly living alone were more likely to be single (never married,divorced,separated and widowed),have lower social support and income,utilize less routine physical check-up and blood glucose screenings,have worse self-reported health and have limitations in IADL compared to the elderly living with other individuals.Women,old age,high education,income and social support and close to medical institutions were positively associated with routine physical check-up among the elderly.Conclusion:Living alone was associated with less preventive care use and worse health.It was important to provide more social support and economic support for the elderly living alone to increase their preventive health care service utilization and improve their health status.

    Relationship between tea dust exposure and lung function: a systemic literature review

    查看更多>>摘要：Background:During the production of tea,tea dust has been viewed as a risk factor that may cause the suffering of changes of lung function and respiratory symptoms.Objective:To review whether the occupational exposure to tea dust may lead to the changes of lung function and respiratory symptoms.Methods:We searched the literatures in March 2012 in the databases of Medline,EMBase,Web of Science,Wan-Fang Database and CNKI.The search condition is “(herb OR tea)AND(lung function OR ventilatory function OR pulmonary function OR spirometric measurement)”.The paper should be pubished in Enghish or Chinese,with internal or external controls.Results:Totally 17 papers were included.Both acute and chronic changes of lung function were referred.In addition,the respiratory symptoms such as cough,tightness and etc.were also reported and discus sed.Conclusion:We concluded that the exposure of tea dust may lead to the changes of lung function and respiratory symptoms.
