<i>In situ</i> Intestinal Absorption Kinetics of Carvedilol Nanosuspensions in Rats

The present study aims to study the in situ intestinal absorption kinetics of carvedilol (CAR) in rats。 The absorption kinetics of CAR solution at different drug concentrations in different segments of intestine with perfusate at different pH values were studied。 The results indicated that drug concentration had little effect on drug absorption。 The pH value of perfusate could significantly affect the drug absorption。 CAR exhibits pH-dependent absorption which can be absorbed more effectively in basic condition。 The absorption of CAR solution and nanosuspension was observed in all parts of the intestinal tract with the main absorption site being jejunum and ileum。 The effective absorption coefficients of CAR nanosuspension in all of the intestinal segments were significantly higher than those of the bulk drug (P < 0。05)。 So we can draw a conclusion that nanosuspension was effective for enhancing intestinal absorption of CAR in rats。
