烟酰胺单核苷酸醇沉干燥工艺研究及微观结构观察

利用烟酰胺单核苷酸微溶于醇类的性质,对醇沉分离纯化烟酰胺单核苷酸的工艺操作条件进行优化,结合真空干燥的方式获得符合标准的烟酰胺单核苷酸.以烟酰胺单核苷酸含量、纯度、收率和水分为指标,考查醇沉温度、醇沉时间等因素,确定烟酰胺单核苷酸的最佳醇沉工艺和干燥工艺,得到烟酰胺单核苷酸的最佳醇沉工艺为取15 g/L的烟酰胺单核苷酸溶液,加入乙醇,使乙醇体积分数达到70%,4℃搅拌1 h,离心得到烟酰胺单核苷酸湿固体,在-0.08 MPa和50℃条件下干燥所得产物的收率最高且水分和纯度达标.利用扫描电镜观察微观结构,分析该工艺制备烟酰胺单核苷酸的可靠性.该优选工艺稳定可靠,可用于烟酰胺单核苷酸的分离纯化.
Using the property that nicotinamide mononucleotide is slightly soluble in alcohols,the process conditions for the separation and purification of nicotinamide mononucleotide by alcohol precipitation were optimized,and the stan-dard nicotinamide mononucleotide was obtained by vacuum drying.The content,purity,yield and moisture of nicotin-amide mononucleotide were used as indicators to investigate the factors such as alcohol precipitation temperature and al-cohol precipitation time.The optimum alcohol precipitation process and drying process of nicotinamide mononucleotide were determined.The optimum alcohol precipitation process of nicotinamide mononucleotide was obtained as follows:15 g/L nicotinamide mononucleotide solution was taken,ethanol was added to make the volume fraction of ethanol reach 70%,stirring at 4℃ for 1 h,and the alcohol precipitate was obtained by filtration.The yield of the product obtained by drying at-0.08 MPa and 50° C was the highest and the moisture and purity were up to standard.The reliability of the preparation of nicotinamide mononucleotide by this process was analyzed by scanning electron microscopy.The opti-mized process is stable and reliable,and can be used for the separation and purification of nicotinamide mononucleotide.
