红曲霉产色素的液态发酵条件优化

目的:通过对液态发酵培养基成分和发酵条件进行优化,提高红曲菌株JTM-1产红曲色素的能力.方法:以分离自红曲米中的一株红曲霉菌JTM-1为实验菌株,采用单因素实验法和正交实验法,优化菌株JTM-1液态发酵培养基.在此基础上,对发酵起始pH、接种量、装液量和发酵温度等条件进行优化.结果:优化得到的最佳培养基条件为大米粉50g/L,蛋白胨14g/L,KCl0.5g/L,K2HPO4 1g/L,MgSO4·7H2O 0.25g/L.最佳发酵条件为发酵起始pH 3.0,接种量8%,装液量25 mU250 mL,发酵温度28℃.在此发酵条件下,发酵液中红色价和黄色价达到最大值,分别为29.68 AU/mL和31.95AU/mL.结论:建立了菌株JTM-1最佳液态发酵培养基和发酵条件,为红曲色素的大规模生产提供了一定的技术支撑.
Optimization of Liquid Fermentation Conditions for Pigment Production of Monascus
Objective:To improve the ability of Monascus JTM-1 to produce pigment by optimizing the composition of liquid fermentation medium and fermentation conditions.Methods:Monascus JTM-1,isolated from red yeast rice,was selected as the experimental strain,and the liquid fermentation medium of Monascus JTM-1 was optimized by single factor experiment and orthogonal experiment.On this basis,the initial pH of fermentation,the amount of inoculation,the amount of liquid filling and the fermentation temperature were optimized.Results:The optimum composition of the fermentation medium was rice flour 50 g/L,peptone 14 g/L,KCl 0.5 g/L,K2HPO4 1 g/L,MgSO4·7H2O 0.25 g/L.The optimum fermentation condi-tions was initial pH 3.0,inoculation amount 8%,medium volume 25 mL/250 mL and fermentation temperature 28℃.Under this fermentation condition,the red and yellow values reached the maximum values,which were 29.68 AU/mL and 31.95 AU/mL,respectively.Conclusion:The optimum liquid fermentation medium and fermentation condition of Monascus JTM-1 were established to provide technical support for the large-scale production of monascus pigment.
