Spherical Nucleic Acid Based on Gold Nanoparticles for Gene Therapy of Brain Tumor
Gene therapy is commonly used to treat brain tumors,but antisense oligonucleotides are difficult to be taken up by cells,which limits the therapeutic effect.In order to improve the therapeutic effect of antisense oligonucleotide(DNA),spherical nucleic acid(Au-DNA)was prepared by binding a large amount of DNA on the surface of gold nanoparticle for tumor gene therapy.Au-DNA was a core-shell structure with a particle size of 100 nm and a negative potential.Due to its special three-dimensional nanostructure,Au-DNA was easier to be taken up by tumor cells.And the cellular uptake effi-ciency of Au-DNA was about four times as high as that of free DNA,which can deliver more DNA into cells.And cell up-take of Au-DNA was time dependent.Due to the high cell uptake efficiency of Au-DNA,the expression level of survivin protein can be reduced,which can effectively inhibit the growth of brain tumor cells.
