摘要

目的:制备共载姜黄素(Cur)/替莫唑胺(TMZ)的脂质纳米粒(lipid nanoparticles,Cur/TMZ-LN).方法:用薄膜分散法、反复冻融法、载体沉积法等制备Cur/TMZ-LN,以包封率为评价指标选出最佳制备方法并进行单因素考查.结果:确定反复冻融法为最佳制备方法,且冻融次数为3,磷脂、胆固醇比为6∶1,磷脂浓度为3mg/Ml,姜黄素包封率为89.23%,替莫唑胺包封率为44.65%.结论:利用反复冻融法制备姜黄素/替莫唑胺脂质纳米粒,包封率较高,粒径较小.

Abstract

Objective:Prepare lipid nanoparticles (Cur/TMZ-LN) co-loaded with curcumin (Cur) and temozolomide (TMZ). Methods:Cur/TMZ-LN was prepared by thin film dispersion,repeated freezing-thawing and carrier deposition. The best preparation method was selected based on the encapsulation efficiency. Results:The method of repeated freez-ing-thawing was confirmed,and the times of freezing-thawing were 3,the ratio of phospholipid to cholesterol was 6∶1,the concentration of phospholipid was 3 mg/mL,the entrapment efficiency of curcumin was 89.23%,the entrapment effi-ciency of temozolomide was 44.65%. Conclusion:Curcumin/temozolomide lipid nanoparticles were prepared by repeated freezing-thawing method with high entrapment efficiency and small particle size.
