Screening,Identification and Optimization of Fermentation Medium for the Production of Polymalic Acid from Aureobasidium Pullulans
A poly malic acid producing strain was selected from natural soil samples and identified as Au-reobasidium pullulans by morphological observation and molecular evolutionary analysis.The selected strain had the ability to synthesize extracellular polymers,such as poly malic acid,pullulan and liamocin. The fermentation medium for PMLA producing was optimized through methods of single factor and Box-Behnken surface response design,and the optimal medium composition was obtained as follows:glu-cose 130 g/L,sodium nitrate 5 g/L,corn steep liquor 1.22 g/L,potassium chloride 0.5 g/L,potassium dihydrogen phosphate 0.1 g/L,magnesium sulfate heptahydrate 0.2 g/L,calcium carbonate 30 g/L.The maximum yield of polymalic acid obtained with this medium was 25.82 g/L,70.72% higher than that before optimization.
