Study on the Tissue Culture and Rapid Propagation Techniques for Acacia mangium x A. auriculiformis
In this paper, new shoots with buds of fine Acacia mangium x A. auriculiformis trees were selected to make such tests as axil-lary bud induction, multiplication and rooting to shorten the multiplication cycle, improve the transplanting survival rate and cultivate robust tissue culture plantlets, laying a good foundation for rapid propagation and promotion of Acacia hybrid while maintaining a high multiplication rate. The results showed that the most suitable disinfection of the explants was 10-second-long soaking in 75%alcohol and 10-min-long treatment in 0.1%mercury chloride, with the survival rate reaching 83%. The best induction medium was MS+6-BA 0.2 mg/L+sucrose 30 g/L, with the germination rate reaching 93.17%. The optimal multiplication medium was MS+6-BA 0.5mg/L+NAA 0.1mg/L+sucrose 30 g/L, with the multiplication times possibly reaching 3.32, and the multiplication cycle shortened to 25d. The optimal rooting medium was MS+IBA 0.5 mg/L+IAA 0.1 mg/L+sucrose 15 g/L, with the rooting rate reaching 91.33%. The sur-vival rate of the transplanting plantlets was 97.8%after 30 days of acclimation.
