大花绣球'史欧尼'的组培快繁技术研究

本文以大花绣球'史欧尼'带腋芽茎段为外植体进行组培快繁技术研究.结果表明:外植体接种于初代完全培养基MS+0.5 mg/L 6-BA+0.02 mg/L NAA+0.1 mg/L GA3+25 g/L蔗糖+6.5~7.5 g/L琼脂中,腋芽萌发时间为 6 d,诱导率高达82%;将不定芽转接至壮苗增殖培养基MS+1.0 mg/L 6-BA+0.02 mg/L NAA+25 g/L蔗糖+6.5~7.5 g/L琼脂中,丛生芽增殖系数可达12.1,且不定芽大多数为健壮芽;在1/2 MS+0.05~0.1 mg/L NAA+0.2~0.3 g/L活性炭培养基上可以很快生根并形成完整植株.
Study on the Tissue Culture and Rapid Propagation Techniques for Hydrangea macrophylla'Masja'
In this paper,a study was made on the tissue culture and rapid propagation techniques for Hydrangea macrophylla'Masja'with stem segments having axillary buds as explants.The results showed that the axillary buds of the explants inoculated on the primary complete medium MS+0.5 mg/L 6-BA+0.02 mg/L NAA+0.1 mg/L GA3+25 g/L sucrose+6.5~7.5 g/L agar germinated in 6 d with an in-duction rate as high as 82%.The cluster buds could reach a multiplication coefficient of 12.1 when the advective buds were transferred to the enrichment medium MS+1.0 mg/L 6-BA+0.02 mg/L NAA+25 g/L sucrose+6.5~7.5 g/L agar,and most of the advective buds were vigorous,which could quickly take root and developed into a complete plant on the medium 1/2 MS+0.05~0.1 mg/L NAA and medium 0.2~0.3 g/L activated carbon.
