Ultraviolet mutagenesis of Morchella esculenta mycelium and screening strains with high yield of polysaccharide
In order to screen for excellent strains of Morchella esculenta, ultraviolet ray was used to treat the mycelium of Morchella. By exploring the growth vigor and polysaccharide content of the mutated mycelium, the mutant strains with fast mycelial growth rate and high polysaccharide content were screened;and the mycelial polysaccharide morphology, infrared spectrum, antioxidant activity and mycelium growth temperature of the mutant strain were analyzed. The results showed that among the 45 screened mutagenic strains, 6 strains with good growth vigor were screened, which had a certain antagonistic effect on the original strain. Among them, the strains of YY-1, YY-2, YY-3, YY-4, and YY-5 had good genetic stability; the mycelium of YY-3, YY-4 and YY-5 strains had high biomass and polysaccharide content. The mycelial polysaccharides of YY-3 strain were flat, with the infrared spectral characteristics of general polysaccharides, and had good ABTS free radical and hydroxyl radical scavenging ability. In addition, the suitable growth temperature for the mycelium of YY-3 strain was 24 ℃. In summary, strains of Morchella with good growth vigor and high polysaccharide content could be selected through ultraviolet mutagenesis screening .
