为了明确大孔树脂脱除穿心莲多糖中色素的工艺参数,研究大孔树脂添加量、脱色时间以及脱色温度对穿心莲多糖脱色效果的影响,采用响应面法优选大孔树脂脱除穿心莲多糖色素的工艺,并探究穿心莲多糖的体外抗氧化活性.结果表明,穿心莲多糖大孔树脂最佳脱色工艺为大孔树脂添加量3.0 g/100 mL多糖溶液,脱色温度50℃,脱色2.0 h,脱色1次.基于该工艺条件,穿心莲多糖色素去除率可达81.35%,多糖保留率73.16%,穿心莲多糖色素去除率和多糖保留率的加权综合评分77.25,表明该穿心莲多糖大孔树脂脱色工艺稳定,可用于穿心莲多糖中色素脱除的生产实际.抗氧化试验结果表明,穿心莲多糖对·OH自由基有较好的清除作用,其具有一定的抗氧化活性.
Optimization of macroporous resin for removing polysaccharide pigment from Andrographis paniculata by response surface method and its antioxidant activity
In order to clarify the process parameters of using macroporous resin to remove pigments from Andrographis paniculata polysaccharides,the effects of resin dosage,decolorization time,and decolorization temperature on the decolorization effect of Andrographis paniculata polysaccharides were studied. The response surface methodology was used to optimize the process of using macroporous resin to remove pigments from Andrographis paniculata polysaccharides,and the antioxidant activity of Andrographis paniculata polysaccharides was investigated. The results showed that the optimal decolorization process for the polysaccharide macroporous resin of Andrographis paniculata was as follows:the amount of macroporous resin added was 3.0 g/100 mL of polysaccharide solution,decolorization temperature was 50℃,decolorization time was 2.0 hours,and decolorization was once. Based on this process condition,the removal rate of pigment from Andrographis paniculata polysaccharide could reach 81.35%,the retention rate of polysaccharides was 73.16%,and the weighted score of Andrographis paniculata polysaccharides pigment removal rate and polysaccharide retention rate was 77.25,which indicated that the macroporous resin decolorization process of Andrographis paniculata polysaccharides was stable and feasible which could be used for the actual production of pigment removal from Andrographis paniculata polysaccharides. The results of antioxidant experiments indicated that the polysaccharides of Andrographis paniculata had a good scavenging effect on·OH free radicals,indicating that the polysaccharides of Andrographis paniculata had certain antioxidant activity.
