Objective:To analyse the expression levels of five target circRNAs (hsa_circ_0008838,hsa_circ_0030716,hsa_circ_0131483,hsa_circ_0053378,and chr1:39927555|39934404) in papillary thyroid carcinoma (PTC) tissues and their correlation with clinical characteristics and to explore its value in the differential diagnosis of PTC and benign nodules (BN).Methods:A total of 30 groups of PTC and BN tissues from the same surgical patient were collected.The expression levels of target circular RNAs were detected by real-time fluorescence quantitative PCR and the differences were analyzed.Group the 30 PTC patients based on different clinicopathological characteristics,compare the differences in expression levels of target circRNAs between groups,and calculate the correlation betw een the expression levels of target circRNAs and preoperative clinical indicators.Receiver Operating Characteristic (ROC) curve was used to analyze the differential diagnostic efficacy of target circRNAs for PTC and BN.Results:In PTC tissues,the expression levels of hsa_circ_0008838,hsa_circ_0030716,hsa_circ_0131483,hsa_circ_0053378 and chr1:39927555|39934404 were higher than those in BN tissues (P<0.05).In PTC tissues with the BRAFV600E mutation,the expression level of hsa_circ_0030716 was lower compared to tissues without the BRAFV600E mutation.In pN1b stage PTC tissues,the expression level of hsa_circ_0131483 was higher than in pN0 and pN1a stages.In multifocal PTC tissues,the expression level of chr1:39927555|39934404 was higher than in solitary PTC tissues.All these differences were significant with P<0.05.Hsa_circ_0053378 showed a positive correlation with TSH,a negative correlation with T3,and hsa_circ_0131483 showed a negative correlation with T4.All these correlations were significant with P<0.05.The AUC values for discriminating between PTC and BN for hsa_circ_0030716,hsa_circ_0008838 and hsa_circ_0131483 were 0.929,0.867 and 0.895,respectively.Conclu sion:The expression of target circRNAs is increased in PTC tissues and correlated with clinical features.hsa_circ_0030716,hsa_circ_0008838 and hsa_circ_0131483 have high differential diagnostic value for PTC and BN.
