Effects of Naoluotong Granule on Apoptosis,PRMT5 and p53 in Rats with Cerebral Ischemia-Reperfusion Injury
Objective:To investigate the effects of Naoluotong Granule on apoptosis,protein arginine methyltrans-ferase 5(PRMT5)and p53 in rats with cerebral ischemia-reperfusion injury.Methods:Adult male SD rats were ran-domly divided into Sham group,model group(MCAO/R)and Naoluotong group(20.2 g/kg).The model group was treated with an improved middle cerebral artery occlusion(MCAO/R)model.Naoluotong group was given Naoluotong granule(20.2 g/kg)once a day for 7 days,and sham operation group and model group were given the same volume of nor-mal saline at the same time by the same method.Neurological function deficit score and infarct volume were observed.Apoptotic rate and protein expression of PRMT5,p53,Bcl-2 and Caspase-3 were detected.Results:Compared with sham operation group,neural function defect was significantly aggravated,cerebral infarction volume was significantly increased,apoptosis rate in brain tissue was significantly increased,PRMT5,p53 and Caspase-3 expressions were significantly increased,and Bcl-2 expression were significantly decreased in model group(P<0.05).Compared with model group,the neurological function defect was significantly reduced,the volume of cerebral infarction was signifi-cantly reduced,the apoptosis rate in brain tissue was significantly decreased,the Bcl-2 expressions were significantly increased,and the expressions of PRMT5,p53 and Caspase-3 were significantly decreased in Naoluotong group(P<0.05).Conclusion:Naoluotong granules can reduce the neurological function deficit score and reduce the volume of cerebral infarction in cerebral ischemia-reperfusion injury rats.Naoluotong granules may play a neuroprotective role in cerebral ischemia-reperfusion injury rats by inhibiting PRMT5 and p53 gene,down-regulating the expression of Cas-pase-3 protein,and up-regulating the expression of Bcl-2 protein to inhibit the cell apoptosis.
