目的:观察通心络胶囊对自发性高血压大鼠(spontaneously hypertensive rat,SHR)的心肌纤维化的干预作用及其机制.方法:8周龄雄性SHR大鼠共40只随机分为通心络高、中、低剂量治疗组及阳性对照组,每组10只;10只8周龄Wistar大鼠作为正常对照组.各组灌胃给药12周后,Masson法检测心肌纤维化,实时逆转录PCR法检测α-SMA和TGF mRNA表达,蛋白印迹检测磷酸化Smad3和CTGF蛋白表达.结果:通心络胶囊减轻了SHR心肌纤维化,抑制 α-SMA mRNA表达,降低了SHR心脏Smad3的磷酸化水平和CTGF的表达,未改变TGFβ1 mRNA的表达.结论:通心络胶囊通过抑制非TGFβ1依赖性Smad3/CTGF促纤维化通路,抑制了心肌成纤维细胞转化,改善了高血压病心肌纤维化.
Intervention Effect of Tongxinluo Capsules on Myocardial Fibrosis in Spontaneously Hypertensive Rats
Objective: To observe the intervention effect and mechanism of Tongxinluo Capsule on myocardial fibro-sis in spontaneously hypertensive rats (SHR). Methods: A total of 408-week-old male SHR rats were randomly divided into high-, medium-, and low-dose Tongxinluo treatment groups and a positive control group, with 10 rats in each group; 108-week-old Wistar rats served as the normal control group. After 12 weeks of oral administration in each group, myocardial fibrosis was detected by Masson's method, α-SMA and TGF mRNA expression were detected by real-time reverse transcription PCR, and phosphorylated Smad3 and CTGF protein expression were detected by West-ern blotting. Results: Tongxinluo capsule alleviated SHR myocardial fibrosis, inhibited α-SMA mRNA expression, reduced the phosphorylation level of Smad3 and CTGF expression in SHR hearts, but did not change the expression of TGFβ1 mRNA. Conclusion: Tongxinluo Capsule inhibits the transformation of cardiac fibroblasts and improves myo-cardial fibrosis in hypertension by inhibiting the TGFβ1-independent Smad3/CTGF pro-fibrotic pathway.
