Effect of Qingshen Granule on Mitochondrial Autophagy in Renal Tissue and Renal Tubular EMT of 5/6 Nephrectomy Rats
Objective: To investigate the effect of Qingshen Granules on mitochondrial autophagy and epithelial cell transdifferentiation (EMT) in renal tissue of 5/6 nephrectomized rats. Method: Thirty male SD rats were randomly di-vided into sham operation group, model group, and Qingshen Granule group, with 10 rats in each group. Except for the sham surgery group, all other 20 rats were made into a 5/6 nephrectomy model. Each group of Qingshen Granules was given corresponding doses of Qingshen Granule aqueous solution; The sham surgery group and model group were given physiological saline by gavage. After continuous administration for 8 weeks, sterile abdominal aortic blood was collected, and 24-hour urine was collected from the metabolic cage. The left kidney was removed. Detect blood and urine creatinine concentrations and calculate endogenous creatinine clearance rate; Western blot method for detect-ing Pink1, Parkin, LC3-Ⅱ α- SMA; Immunofluorescence assay was used to detect the co localization expression of LC3-Ⅱ and mitochondrial membrane protein VDAC1 in renal tissue; HE and Masson staining were used to observe the pathological changes of rat kidneys under light microscopy; Transmission electron microscopy was used to exam-ine the ultrastructure of mitochondria in renal tubular epithelial cells.Result: Model group rat.Conclusion: Qingshen Granules can upregulate the expression of miR-23b-5p in NRK-52E cells and inhibit the process of NRK-52E cell transdifferentiation by enhancing the mitochondrial autophagy activity mediated by the PINK1/Parkin pathway.
