摘要
目的:探讨淫羊藿总黄酮(TFE)对去势大鼠抗酒石酸酸性磷酸酶5b(TRACP-5b)及骨密度的影响.方法:10月龄雌性SD大鼠去卵巢法建立大鼠模型,随机数字表法分为空白对照组(N)、模型对照组(O)、TFE低(TL)、中(TM)、高(TH)剂量组,β-雌二醇(E)组各10只,灌胃干预90d.用抗酒石酸酸性磷酸酶试剂盒测定TRACP-5b含量,用小动物分析软件测定活体全身骨密度.结果:①TRACP-5b水平O组明显高于N组,具有显著性差异(P<0.01),TL、TM、TH和E组TRACP-5b水平明显低于O组,具有显著性差异(P<0.01).②与N组比较,O、TL、TM、TH和E组腰椎骨密度均下降,唯有O组呈显著性差异(P<0.05);与O组比较,TM、TH组腰椎骨密度升高、呈显著性差异(P<0.05),E组腰椎骨密度升高、具有显著性差异(P<0.01).③与N组比较,O、TL、TM、TH和E组全身骨密度均下降,唯有O组差异有统计学意义(P<0.05);与O组比较,TL、TM、TH和E组全身骨密度均升高,唯有E组呈显著性差异(P<0.05).结论:TFE可以降低去势大鼠血清中TRACP-5b的含量,并与去势大鼠腰椎骨密度、全身骨密度呈一定的量效关系.
Abstract
Objective: To explore the effect of epimedium total flavonoids (TFE) on tartrate-resistant acid phos-phatase 5b (TRACP-5b) and bone mineral density in ovariectomized rats. Methods: 10-month-old female SD rats were ovariectomized to establish a rat model. The random number table method was divided into blank control group (N), model control group (O), TFE low (TL), medium (TM), and high dose. Group (TH) and β-estradiol group (E), 10 ani-mals each, were administered intragastric intervention for 90 days. The tartrate-resistant acid phosphatase kit was used to determine the TRACP5b content, and the small animal analysis software was used to determine the whole body bone mineral density in vivo. Results:① The level of TRACP-5b in group O was significantly higher than that in group N, with a significant difference (P<0.01). The levels of TRACP-5b in groups TL, TM, TH and E were significantly lower than that in group O, with a significant difference (P<0.01). ② Compared with group N, the bone density of lumbar vertebrae in groups O, TL, TM, TH and E all decreased, and only group O showed a significant difference (P<0.05);compared with group O, the bone density of lumbar vertebrae in groups TM and TH decreased. The bone density of the lumbar vertebrae in group E increased with a significant difference (P<0.05). The bone density of the lumbar spine in group E increased with a significant difference (P<0.01). ③ Compared with the N group, the whole body bone density of the O, TL, TM, TH and E groups all decreased, and only the difference in the O group was statistically significant (P<0.05); compared with the O group, the TL, TM, TH The whole body bone density of group E and group E increased, but there was a significant difference only in group E (P<0.05). Conclusion: TFE can reduce the content of TRACP-5b in the serum of ovariectomized rats, and has a certain dose-effect relationship with the lumbar spine bone density and whole body bone density of ovariectomized rats.
