Exploring the Action Mechanism of the Shenfu Decoction on Chronic Heart Failure Based on Bioinformatics
Objective:To explore the active ingredients,genes and signaling pathways of Shenfu Decoction in treat-ing chronic heart failure(CHF)through bioinformatics methods and explore its potential mechanism.Methods:The TCMSP database was used to search for the active ingredients and targets of traditional Chinese medicine contained in Shenfu Decoction.The chip data set of chronic heart failure was screened out through the GEO database and differential analysis was performed.The related genes of CHF were obtained and then compared with the targets of traditional Chi-nese medicine.The target genes were obtained through intersection;the cytoscape3.8.2 software was used to construct the active ingredient-target gene network and screen out the important active ingredients;finally,a protein interaction network of the target genes was constructed and the target genes were enriched with GO biological processes and KEGG pathways.Set analysis.Results:25 active ingredients were screened out from Shenfu Decoction,and there were 8 com-mon target genes related to CHF.The important active ingredients were kaempferol,ginsenoside rh2,and β-sitosterol;the key target genes Including PTGS2,IL1B,JUN,NFKBIA,CASP1,etc.;GO biological process results show that it is related to the response to mechanical stimulation,fever,response to lipopolysaccharide,etc.;KEGG pathway enrich-ment analysis results show that it mainly involves C-type lectin receptor signaling pathway,IL-17 signaling pathway and TNF signaling pathway.Conclusion:This study analyzed the potential mechanism of action of Shenfu Decoction in treating CHF through bioinformatics,and initially clarified that Shenfu Decoction has the characteristics of multi-component,multi-target and multi-pathway involvement in treating CHF,providing a theoretical basis for subsequent in-depth research.
