Study on the Mechanism of Action of"Pueraria lobata-Mulberry leaf"Medicine in the Treatment of Diabetic Nephropathy
Objective:To explore the mechanism of action of"Pueraria lobata-Mulberry leaf"medicine in the treatment of diabetic nephropathy by using the method of network pharmacology.Methods:First,the TCMSP platform was used to screen out the active ingredients and targets of the"Pueraria lobata-Mulberry leaf"drug pair,and a to-tal of 5 databases including GeneCards,DrugBank,OMIM,PharmGkb,and TTD were used to screen out target genes related to diabetic kidney disease.Use R language to obtain the common targets of drugs and diseases,draw a Venn diagram,and use Cytoscapes software to construct and analyze the drug component-target regulatory network.Use the STRING database platform to construct PPI networks and screen core targets.GO function and KEGG pathway enrich-ment analysis of key targets was performed using R language.Results:The"Pueraria lobata-Mulberry leaf"drug pair has a total of 33 effective active ingredients and 52 targets.The beta-sitosterol active ingredient exists in the traditional Chinese medicine Pueraria lobata and mulberry leaf.There are 167 common targets between drug pairs and diabetic nephropathy,including 48 key targets.The PPI network of intersection genes between drugs and diseases contains 618 edges and 167 nodes,and its core genes include GSK3B,PGR,HSP90AA1,ESR1,etc.Through GO enrichment analy-sis,strip enrichment results were obtained,and the form and function of genes were analyzed from three levels:BP,CC,and MF.KEGG enrichment analysis(P<0.01)obtained 30 relevant signaling pathways.Based on the analysis of the top 30 P-values,the main pathways include neurodegeneration pathways-multiple disease signaling pathways,PI3K-Akt signaling pathways,blood lipids and Atherosclerosis signaling pathways,etc.Conclusion:"Pueraria lobata-mulberry leaf"medicine plays a role in treating diabetic kidney disease by participating in biological processes such as inflam-matory response and oxidative stress.
