Study on the Effect of Gandou Bushen Decoction on the Hypothalamus-pituitary Reproductive Endocrine in Female Wilson Disease Model TX Mice
Objective:To investigate the effect of Gandou Bushen Decoction on the reproductive endocrine of hy-pothalamus-pituitary in female Wilson's disease TX mice.Methods:TX mice and normal control mice were randomly divided into control group,Wilson's disease model group and Gandou Bushen Decoction group,10 mice in each group.The pathological morphology and ultra-structure of hypothalamus and pituitary tissues in female Wilson's disease TX mice were observed.The levels of GnRH,FSH,LH and E2 in serum of mice were detected by ELISA method,the pathological morphology and structural changes of hypothalamus and pituitary tissues of mice were observed by hema-toxylin-eosin staining method,the ultra-structure of mouse gonadotropin cells was observed by transmission electron microscopy,the expression of GnRH protein in mouse hypothalamus tissue was detected by immunohistochemistry,and its average optical density level was determined.The pregnancy rate and litter size of mice were observed.Results:Compared with the normal control group,the levels of GnRH,FSH,LH,and E2 in the serum of the model group TX mice were decreased(P<0.05),the number of neurons in the hypothalamus and pituitary decreased,a large number of neurons showed nuclear condensation,blurred cell boundaries,severe pathological changes,and decreased pregnancy rate and litter birth rate;compared with the model group,the levels of GnRH,FSH,LH,and E2 in the serum of the mice in the Gandou Bushen Decoction group were increased(P<0.05),the number of hypothalamic neurons increased compared with the model group,the edema and vacuolar changes of pituitary tissue cells decreased,the expression of GnRH protein increased(P<0.05),and the pregnancy rate and litter birth rate increased.Conclusion:Gandou Bushen Decoction can improve the secretion level of reproductive hormones and up-regulate the expression level of GnRH protein through the regulation of hypothalamus-pituitary on reproductive function,thereby improving the reproductive damage of female Wilson's disease TX mice.
