Mechanism of Migration,Invasion and Glycolysis of Human Colon Cancer Cells Induced by Diyuhuaihua Decoction Via KRT17 Gene
AIM:To explore the effects of Diyuhuaihua Decoction on human colon cancer cell viability,invasion and migration ability and glycolysis by silencing KRT17 gene expression METHODS:Human colon cancer HCT116 cells cultured in vitro were treated with a blank control group and low,medium and high(10%,15%,20%)vol-ume serum containing Diyuwuhua decoction.The cell viability was detected by CCK8 method,migration and invasion were detected by Transwell,glucose and lactic acid levels were detected by glycolysis kit.The differentially expressed gene KRT17 in colon cancer was screened by GEO and GEPIA databases,to detect the effect of Diyuhuahua decoc-tion on KRT17 and effect of KRT17 on viability,migration,invasion and glycolysis of colon cancer cells.The effects of Diyuhuaihua Decoction/KRT17 axis on cell viability,migration,invasion and glycolysis were detected.RESULTS:Diyuhuaihua decoction can inhibit the activity,migration,invasion and glycolysis of colon cancer cells(P<0.05).The KRT17 gene is highly expressed in colon cancer and can promote the viability,migration,invasion and glycolysis of colon cancer cells(P<0.05).Diyuhuaihua decoction can inhibit the expression of KRT17(P<0.05),and overex-pression of KRT17 can reverse the inhibitory effect of Diyuhuaihua decoction on the viability,migration,invasion and glycolysis of colon cancer cells(P<0.05).CONCLUSION:Diyuhuaihua decoction can inhibit the viability,migration and invasion of colon cancer cells and promote glycolysis by regulating the expression of KRT17 gene
