Study on the Effect of Zhongwan Acupoint Combined with Shenxiangsan Application on Gastroesophageal Reflux Via SCF/C-kit Signaling Pathway
Objective:To investigate the regulatory effect of Tuizhongwan combined with Shenxiang powder appli-cation on gastroesophageal reflux(GER)through stem cell factor(SCF)/C-kit signaling pathway.Methods:GER ani-mal models were established by dietary indiscriminateness and catheter balloon dilatation.GER rabbits were treated with Tuizhongwan,Shenxiang powder and Tuizhongwan combined with Shenxiang powder application,respectively.Pathological damage of gastric antrum tissue was observed by hematoxylin-eosin staining.Immunofluorescence was used to detect the fluorescence intensity of C-kit.RT-qPCR was used to detect the relative expression of SCF and C-kit mRNA.Western blot was used to detect the protein expression of SCF,C-kit and connexin 43(CX43).Results:In the GER group,cellular inflammatory infiltration was obvious in the gastric antrum tissue of rabbits,gastric mucosal epithelial cells proliferated,and the expression of SCF,C-kit and CX43 in the tissue was significantly reduced.Tu-izhongwan and Shenxiang powder application can reduce the degree of inflammatory infiltration in gastric antrum tissue and promote the expression of SCF,C-kit and CX43.Compared with the Tuizhongwan or Shenxiang powder application alone group,the Tuizhongwan combined with Shenxiangsan application group showed a weakened cellular inflamma-tory infiltration,almost no cell proliferation,and significantly increased SCF,C-kit and CX43 expression.Conclusion:Tuizhongwan combined with Shenxiang powder application can significantly improve GER,which may be achieved by regulating the SCF/C-kit signaling pathway.
