摘要
目的:探讨黄地安消胶囊对肾阴虚型糖尿病大鼠炎症的影响.方法:70只大鼠随机分为空白组、模型组、黄地安消胶囊低剂量组、黄地安消胶囊中剂量组、黄地安消胶囊高剂量组、二甲双胍组和中药对照组.ELISA检测白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、瘦素(Leptin)和胰高糖素样肽-1(GLP-1)的浓度水平,RT-qPCR和WB实验分别检测mRNA表达水平和蛋白表达水平.结果:与空白组相比,模型组大鼠血清和胰腺组织中Leptin、IL-1β、IL-6和TNF-α表达水平均显著上升,而GLP-1水平下降.黄地安消高剂量组、二甲双胍组和中药对照组相较于模型组的大鼠血清和胰腺组织中Leptin、IL-1β、IL-6和TNF-α表达水平均显著下降,而GLP-1表达水平显著上升.结论:黄地安消胶囊可通过抑制肾阴虚型糖尿病大鼠炎症状态发挥治疗作用.
Abstract
Objective:Exploring the effect of Huangdi Anxiao Capsules on inflammation in kidney-yin-deficient diabet-ic rats. Methods:Seventy rats were randomly divided into a blank group,a model group,a low-dose group of Huangdi Anxiao Capsules,a medium-dose group of Huangdi Anxiao Capsules,a high-dose group of Huangdi Anxiao Capsules,a metformin group,and a Traditional Chinese medicine control group.The concentration levels of GLP-1,Leptin,IL-1β,IL-6 and TNF-α were detected by ELISA,and the levels of mRNA and protein expression were detected by RT-qPCR and Western blot. Results:Compared with the blank group,the expression levels of Leptin,IL-1β,IL-6 and TNF-α were significantly increased in serum and pancreatic tissues of rats in the model group,while the level of GLP-1 was decreased. The factors,such as Leptin,IL-1β,IL-6 and TNF-α,were significantly decreased in serum and pancre-atic tissues of the rats in the high dose group of Huangdi Anxiao Capsules compared with the model group,while the level of GLP-1 was increased. Conclusion:Huangdi Anxiao Capsules may exert therapeutic effects by inhibiting the inflammatory state of kidney-yin-deficient diabetic rats.
