Study on the Effect of Danggui Shaoyao San on Vascular Dementia based on Glutamate and N-methyl-D-Aspartate Receptors in Hippocampal Tissue
Objective:To investigate the effect of Danggui Shaoyao San(DSS)on vascular dementia(VD)via gluta-mate and N-methyl-D-aspartate receptors(NMDAR)in hippocampal tissue.Methods:VD rats were modeled by bi-lateral carotid artery permanent ligation.The rats with successful modeling were randomly divided into 5 groups:sham operation group,model group,DSS low-dose group,DSS high-dose group,and positive control group.The learning and memory ability of rats was tested by Morris water maze;the damage of neurons in the hippocampus of rats was observed by HE staining;and the changes in the contents of NMDAR and Caspase-3 were determined by immunohistochemistry.Results:Compared with the sham operation group,the escape latency of the model group was significantly prolonged(P<0.01),and the difference was statistically significant.Compared with the model group,the escape latency of the low-dose DSS group,high-dose DSS group and nimodipine group was shortened(P<0.01);compared with the sham operation group,the NMDAR and Caspase-3 of the model group were significantly increased(P<0.01).Compared with the model group,the NMDAR and Caspase-3 of the high-dose DSS group were significantly decreased in the low-dose DSS group,high-dose DSS group and nimodipine group(P<0.01).Conclusion:DSS can significantly improve the learning and memory ability of VD rats,and may have a therapeutic effect on VD through hippocampal glutamate and NMDAR.
