目的:探讨当归芍药散(DSS)通过海马组织谷氨酸、N-甲基-D-天冬氨酸受体(NMDAR)对血管性痴呆(VD)影响的研究.方法:VD大鼠的造模采用的双侧颈动脉永久结扎法,将造模成功大鼠随机分为5组,假手术组、模型组、DSS低剂量组、DSS高剂量组、阳性对照组.大鼠的学习记忆能力通过Morris水迷宫来进行检测;大鼠海马组织神经元受损情况通过HE 染色来观察;采用免疫组化方法测定NMDAR、Caspase-3的含量变化.结果:与假手术组比较,模型组逃避潜伏期显著延长(P<0.01),差异具有统计学意义.与模型组比较,DSS低、高剂量组与尼莫地平组逃避潜伏期缩短(P<0.01);与假手术组比较,模型组的NMDAR、Caspase-3 明显升高(P<0.01).DSS低剂量组、DSS高剂量组、尼莫地平组与模型组比较,DSS高剂量组NMDAR、Caspase-3 明显降低(P<0.01).结论:DSS能明显改善VD大鼠的学习记忆能力,可能通过海马组织谷氨酸、NMDAR对VD起治疗作用.
Study on the Effect of Danggui Shaoyao San on Vascular Dementia based on Glutamate and N-methyl-D-Aspartate Receptors in Hippocampal Tissue
Objective:To investigate the effect of Danggui Shaoyao San(DSS)on vascular dementia(VD)via gluta-mate and N-methyl-D-aspartate receptors(NMDAR)in hippocampal tissue.Methods:VD rats were modeled by bi-lateral carotid artery permanent ligation.The rats with successful modeling were randomly divided into 5 groups:sham operation group,model group,DSS low-dose group,DSS high-dose group,and positive control group.The learning and memory ability of rats was tested by Morris water maze;the damage of neurons in the hippocampus of rats was observed by HE staining;and the changes in the contents of NMDAR and Caspase-3 were determined by immunohistochemistry.Results:Compared with the sham operation group,the escape latency of the model group was significantly prolonged(P<0.01),and the difference was statistically significant.Compared with the model group,the escape latency of the low-dose DSS group,high-dose DSS group and nimodipine group was shortened(P<0.01);compared with the sham operation group,the NMDAR and Caspase-3 of the model group were significantly increased(P<0.01).Compared with the model group,the NMDAR and Caspase-3 of the high-dose DSS group were significantly decreased in the low-dose DSS group,high-dose DSS group and nimodipine group(P<0.01).Conclusion:DSS can significantly improve the learning and memory ability of VD rats,and may have a therapeutic effect on VD through hippocampal glutamate and NMDAR.
