基于AMPK通路探究电针结合有氧运动对肥胖大鼠的作用机制

目的 观察电针结合有氧运动对肥胖大鼠的影响,并初步探究其作用机制.方法 从45只SD大鼠中随机挑选8只作为空白组,其余大鼠用高脂饲料喂养8周建立肥胖大鼠模型,剔除模型复制失败的5只大鼠,将模型复制成功的大鼠分为模型组、电针组、运动组、电针联合运动组,每组8只;除空白组和模型组外,电针组采用电针、运动组采用运动、电针联合运动组采用电针联合运动措施干预8周.监测各组大鼠体质量,采用苏木精—伊红染色观察大鼠肝脏组织形态变化,采用ELISA法检测大鼠血清总胆固醇(total cholesterol,TC)、三酰甘油(triglyceride,TG)、低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)、高密度脂蛋白胆固醇(high-density lipo-protein cholesterol,HDL-C)、白细胞介素(interleukin,IL)-1β、IL-6、三磷酸腺苷(adenosine-triphosphate,ATP)水平,Western blot法检测大鼠肝组织磷酸化肝激酶B1(phosphorylated liver kinase B1,p-LKB1)、磷酸化腺苷酸活化蛋白激酶(phosphorylated adenylate-activated protein kinase,p-AMPK)、乙酰辅酶 A 羧化酶(acetyl Co A carboxyl-ase,ACC)蛋白表达水平.结果 与模型组比较,电针结合运动组可以显著降低大鼠体质量,血清TC、TG、HDL-C、IL-1β、IL-6水平,肝组织ACC蛋白表达水平(P<0.05);显著增加血清HDL-C、ATP水平,肝组织p-LKB1、p-AMPK蛋白表达水平(P<0.05).结论 电针联合运动可改善肥胖大鼠脂质能量代谢,减少炎症反应,其机制可能与激活AMPK信号通路有关.
Mechanism of Action of Acupuncture Combined with Aerobic Exercise in Obese Rats:A Study Based on the AMPK Pathway
Objective To investigate the effect of acupuncture combined with aerobic exercise on obese rats and its mechanism of action.Methods Among 45 Sprague-Dawley rats,8 were randomly selected as blank group,and the rest of the rats were fed with high-fat diet for 8 weeks to establish a rat model of obesity.Five rats with unsuccessful modeling were removed,and the rats with successful modeling were divided into model group,acupuncture group,exercise group,and acupuncture-exercise group,with 8 rats in each group.All rats except those in the blank group and the model group were given acupuncture and exer-cise intervention measures for 8 weeks.Body weight was monitored for the rats in each group;HE staining was used to observe liver histomorphological changes;ELISA was used to measure the serum levels of total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C),interleukin-1β(IL-1β),interleukin-6(IL-6),and adenosine triphosphate(ATP);Western blot was used to measure the protein expression levels of phosphorylated liver kinase B1(p-LKB1),phosphorylated adenylate-activated protein kinase(p-AMPK),and acetyl-CoA carboxylase(ACC)in rat liver tissue.Results Compared with the model group,the acupuncture-exercise group had significant reductions in body weight,the serum levels of TC,TG,HDL-C,IL-1β,and IL-6,and the protein expression level of ACC in liver tissue(P<0.05)and significant increases in the serum levels of HDL-C and ATP and the protein expression levels p-LKB1 and p-AMPK in liver tissue(P<0.05).Conclusion Acupuncture combined with exercise can improve lipid-energy metabolism and reduce inflamma-tory response in obese rats,possibly by activating the AMPK signaling pathway.
