电针对局灶性脑缺血大鼠JAK2/STAT3通路、血管内皮生长因子的影响及其对神经保护作用机制的研究

目的 观察电针"百会""大椎"对局灶性脑缺血大鼠酪氨酸蛋白激酶2/信号转导和转录激活因子3(Janus kinase 2/signal transducer and activator of transcription 3,JAK2/STAT3)通路和血管内皮生长因子(vascular endo-thelial growth factor,VEGF)的影响,探讨电针对局灶性脑缺血大鼠神经保护的作用机制.方法 将36只SD大鼠随机分为正常组、假手术组、模型组、电针组,每组9只,采用Longa线栓法进行局灶性脑缺血模型复制,电针组模型复制4 h后予以电针治疗,共治疗7 d.比较各组大鼠模型复制后4 h和7 d的神经功能评分;Western blot法检测各组大鼠脑组织JAK2、磷酸化酪氨酸蛋白激酶2(phosphorylated Janus kinase 2,p-JAK2)、STAT3、磷酸化信号转导和转录激活因子3(phosphorylated signal transducer and activator of transcription 3,p-STAT3)、VEGF蛋白表达水平;RT-PCR法检测各组大鼠脑组织JAK2、STAT3、VEGF mRNA表达水平.结果 模型复制后4 h,电针组、模型组大鼠神经功能评分显著高于假手术组和正常组(P<0.05);与假手术组比较,模型组JAK2、p-JAK2、STAT3、p-STAT3、VEGF蛋白表达水平均显著升高(P<0.05),蛋白表达水平显著升高(P<0.05);JAK2、STAT3、VEGF mRNA表达水平显著升高(P<0.05);与模型组比较,电针组7 d后神经功能评分显著降低(P<0.05);STAT3、VEGF、p-STAT3、JAK2、p-JAK2蛋白表达水平显著升高(P<0.05);JAK2、STAT3、VEGF mRNA表达水平显著升高(P<0.05).结论 电针可能通过激活JAK2/STAT3通路、促进下游VEGF的表达发挥对局灶性脑缺血大鼠的神经保护作用.
Effect of Electroacupuncture on the Janus Kinase-2/Signal Transducer and Activator of Transcription 3 Pathway and Vascular Endothelial Growth Factor and Its Neuroprotective Mechanism in Rats with Focal Cerebral Ischemia
Objective To investigate the effect of electroacupuncture at Baihui and Dazhui points on the Janus kinase 2 (JAK2)/signal transducer and activator of transcription 3 (STAT3)pathway and vascular endothelial growth factor (VEGF),as well as its neuroprotective mechanism in rats with focal cerebral ischemia.Methods A total of 36 Sprague-Dawley rats were randomly divided into normal group,sham-operation group,model group,and electroacupuncture group,with 9 rats in each group.The Longa suture method was used to establish a model of focal cerebral ischemia,and the rats in the electroacupuncture group were given electroacupuncture at 4 hours after modeling,for 7 days in total.After modeling,the groups were compared in terms of neurological deficit score at 4 hours and 7 days after surgery;Western blot was used to measure the protein expression levels of JAK2,phosphorylated JAK2 (p-JAK2),STAT3,phosphorylated STAT3 (p-STAT3),and VEGF in brain tissue,and RT-PCR was used to measure the mRNA expression levels ofJAK2,STAT3,and VEGF in brain tissue.Results At 4 hours after modeling,the electroacupuncture group and the model group had a significantly higher neurological deficit score than the sham-group and the normal group (P<0.05).Compared with the sham-operation group,the model group had significant increases in the protein expression levels of JAK2,p-JAK2,STAT3,p-STAT3 and VEGF (P<0.05)and the mRNA expression levels of JAK2,STAT3,and VEGF (P<0.05).Compared with the model group,the electroacupuncture group had a significant re-duction in neurological deficit score after 7 days (P<0.05)and significant increases in the protein expression levels of STAT3, VEGF,p-STAT3,JAK2,and p-JAK2 (P<0.05)and the mRNA expression levels of JAK2,STAT3,and VEGF (P<0.05).Conclusion Electroacupuncture exerts a neuroprotective effect on rats with focal cerebral ischemia possibly by activa-ting the JAK2/STAT3 pathway and promoting the expression of downstream VEGF.
