Astragalus Polysaccharides Inhibit the Growth of Ovarian Adenocarcinoma Caov-3 Cells by Regulating the Bcl-2/Bax Signaling Pathway:An Experimental Study
Objective To investigate the effect of Astragalus polysaccharides on the growth of ovarian adenocarcinoma Caov-3 cells by regulating the Bcl-2/Bax signaling pathway.Methods Caov-3 cells were divided into low-,middle-,and high-dose groups and were added with Astragalus polysaccharides at a concentration of 10,50,and 100 mg/mL,respectively.Flow cy-tometry was used to observe the effect on cell cycle after Caov-3 cells were treated with Astragalus polysaccharides at a concen-tration of 10,50,and 100 mg/mL,respectively;microscopy was performed to observe the apoptosis and morphological chan-ges of Caov-3 cells;Western blot was used to observe the effect of Astragalus polysaccharides on the protein expression levels of Bcl-2 and Bax and Bcl-2/Bax ratio.Meanwhile,the inhibition rate of Caov-3 cells was compared between aspirin combined with Astragalus polysaccharides and Astragalus polysaccharides used alone.Results The inhibition rates of Caov-3 cells in the low-,middle-,and high-dose Astragalus polysaccharides groups were 40%,50%,and 65%,respectively,and the half-maxi-mal inhibitory concentration of Astragalus polysaccharides was 50 mg/mL.Compared with the control group,the high-dose Astragalus polysaccharides group had the most significant inhibitory effect on Caov-3 cells (P<0.05),and Astragalus poly-saccharides combined with aspirin had a significantly better inhibitory effect than Astragalus polysaccharides alone (P<0.05). Compared with the control group,the high-dose Astragalus polysaccharides group had a significantly greater effect on the cell cycle of Caov-3 cells (P<0.05). Compared with the control group,the high-dose Astragalus polysaccharides group had a sig-nificant increase in the number of apoptotic Caov-3 cells,a significant reduction in the protein expression level of Bcl-2 and Bcl-2/Bax ratio,and significant increases in the protein expression level of Bax(P<0.05). Conclusion Astragalus polysac-charides can effectively inhibit the growth of Caov-3 cells and has a better effect when combined with aspirin,possibly by regu-lating the Bcl-2/Bax signaling pathway and promoting cell apoptosis.
