基于肠道菌群探究艾灸"百会"改善大鼠抑郁状态的作用机制

目的 通过对大鼠肠道菌群进行16S rRNA测序,探究艾灸"百会"对抑郁样大鼠肠道菌群的影响,并分析迷走神经在艾灸治疗抑郁样大鼠中所发挥的作用.方法 将大鼠随机分为空白组、模型组、假手术艾灸组、手术艾灸组.采用慢性不可预见性轻度应激与孤养结合的方法复制抑郁状态大鼠模型.模型复制成功后,暴露但不切断假手术艾灸组大鼠迷走神经,切断手术艾灸组大鼠迷走神经,术后给予艾灸治疗21 d.模型复制前后及末次艾灸后均对大鼠进行行为学观察;末次艾灸后检测各组大鼠海马和结肠组织中脑源性神经营养因子(brain-derived neu-rotrophic factor,BDNF)水平;观察各组大鼠粪便中肠道菌群变化.结果 与模型组比较,手术艾灸组及假手术艾灸组大鼠体质量,旷场实验运动速度,1%糖水消耗率,海马、结肠组织BDNF蛋白表达水平均显著升高(P<0.05),旷场实验静止时间显著缩短(P<0.05).模型组、假手术艾灸组及手术艾灸组大鼠肠道菌群结构从门水平至属水平均发生改变,发生变化的主要菌群为螺旋体门、乳酸菌科、毛螺菌科及普罗沃氏菌科等菌群.4组大鼠肠道菌群多样性发生改变,空白组大鼠肠道菌群多样性与其余3组存在显著差异,其中假手术艾灸组α多样性指数最高.结论 切断迷走神经可增强艾灸"百会"升高机体BDNF表达水平的作用;艾灸"百会"可提高肠道菌群多样性,维护肠道菌群稳态;切断迷走神经可降低艾灸"百会"对肠道菌群的影响,使肠道菌群多样性降低.
Mechanism of Action of Moxibustion at Baihui Point in Improving Depressive State in Rats:A Study Based on Intestinal Flora
Objective To investigate the effect of moxibustion at Baihui point on intestinal flora in depression-like rats through 16S rRNA sequencing of intestinal flora,as well as the role of the vagus nerve in moxibustion treatment of depression-like rats.Methods Rats were randomly divided into control group,model group,surgery+moxibustion group,and sham-operation+moxibustion group.Chronic unpredictable mild stress was used to establish a model of depressive state.After successful mod-eling,the vagus nerve was exposed without being cut off for the rats in the sham-operation+moxibustion group,while the va-gus nerve was cut off for those in the surgery+moxibustion group,followed by moxibustion treatment for 21 days.Behavioral observation was performed for the rats before modeling,after modeling,and after the last session of moxibustion treatment;the level of brain-derived neurotrophic factor (BDNF)in hippocampal tissue and colon tissue was measured after the last session of moxibustion treatment,and the change in fecal intestinal flora was observed for each group.Results Compared with the model group,the surgery+moxibustion group and the sham-operation+moxibustion group had significant increases in body weight,speed of movement in the open field test,1% sugar water consumption,and the protein expression level of BDNF in hippocampal tissue and colon tissue (P<0.05)and a significant reduction in rest time in the open field test (P<0.05).The model group,the sham-operation+moxibustion group,and the surgery+moxibustion group had changes in the composition of intestinal flora from the phylum level to the genus level,and such changes mainly involved Spirochaete,Lactobacillaceae,Lach-nospiraceae,and Prevotellaceae.All four groups showed changes in the diversity of intestinal flora,with a significant difference between the blank group and the other three groups,and the sham-operation+moxibustion group had the highest α-diversity index.Conclusion Severing the vagus nerve can help to enhance the effect of moxibustion at Baihui point in upregulating the level of BDNF in the body,and moxibustion at Baihui point can improve the diversity of intestinal flora and maintain the homeo-stasis of intestinal flora.Severing the vagus nerve can weaken the effect of moxibustion at Baihui point on intestinal flora and reduce the diversity of intestinal flora.
