Determination of Tetracycline and Oxytetracycline in Sheep Milk by SPE-HPLC
A method for simultaneous determination of tetracycline and oxytetracycline in sheep milk using solid-phase extraction-high-performance liquid chromatography was established.The analytes were extracted with 0.1 mol·L-1 EDTA Mcllvaine buffer,purified with HLB solid-phase extraction column,eluted with formic acid-methanol solution,and the concentration of the analytes were determined by HPLC.The results showed that tetracycline and oxytetracycline had a good linear relationship in the ranges from 0.1 to 100 μg·mL-1,and the detection limits of tetracycline and oxytetracycline were 0.021 μg·mL-1 and 0.030 µg·mL-1,respec-tively.The recovery rates of tetracycline and oxytetracycline were 95.15%~95.94%and 89.84%~97.93%,respectively.This method was accurate,reliable,and has good repeatability,which can be used for the determi-nation of tetracycline and oxytetracycline in sheep milk.
