Diagnostic value of miRNA-182,miRNA-200c,and pepsinogen Ⅰ/pepsinogen Ⅱ single and combined detection in gastric cancer
Objective To explore the diagnostic value of miRNA-182,miRNA-200c,and pepsinogen Ⅰ(PG Ⅰ)/pep-sinogen Ⅱ(PG Ⅱ)single and combined detection in gastric cancer.Method A total of 65 gastric cancer patients and 61 healthy subjects were selected as observation group and control group.Enzyme-linked immunosorbent assay(ELISA)was used to detect serum levels of PG Ⅰ and PG Ⅱ in both groups.Reverse transcription-polymerase chain reaction(RT-PCR)was used to detect levels of miRNA-182,miRNA-200c in both groups. The receiver operating characteristic(ROC)curve was drawn and the area under the curve(AUC)was calculated to evaluate the diagnostic value of miRNA-182,miRNA-200c,and PG Ⅰ/PG Ⅱ single and combined detection for gastric cancer.Result The serum levels of miRNA-182,miRNA-200c in the observation group were higher than those in the control group, and the differences were sta-tistically significant(P<0.01).The serum levels of PG Ⅰ and PG Ⅰ/PG Ⅱ in the observation group were lower than those in the control group, while the level of PG Ⅱ was higher than that in the control group, and the differences were statistical-ly significant(P<0.05).The ROC curve showed that the AUC for the combined detection of miRNA-182,miRNA-200c,and PG Ⅰ/PG Ⅱ in the diagnosis of gastric cancer was 0.829(95%CI:0.633-0.859),which was higher than the single detec-tion of 0.650,0.722,and 0.818 for each indicator. At this time, the sensitivity was 0.825,and the specificity was 0.890.Conclusion The combined detection of miRNA-182,miRNA-200c,and PG Ⅰ/PG Ⅱ has higher diagnostic value for gas-tric cancer, and may serve as an effective indicator for gastric cancer diagnosis.
