Expression of hexokinase 2 in glioma and its relationship with prognosis
Objective To investigate the expression of hexokinase 2(HK2)in glioma and its relationship with prog-nosis.Method The data sets of HK2 mRNA expression in glioma and the clinical data of corresponding patients were downloaded from The Cancer Genome Atlas(TCGA)database and the Chinese Glioma Genome Atlas(CGGA)database.The expressions of HK2 mRNA in glioma patients with different World Health Organization(WHO)grades,gender,nes-tin(NES)mRNA expression,isocitrate dehydrogenase(IDH)mutation status,chromosome 1p/19q co-deletion status,and ATRX mutation status were compared.Using UCSC Xena and other online tool websites,the Kaplan-Meier method was used to draw the survival curves of the HK2 mRNA high expression group and the low expression group.The expression levels of HK2 mRNA were combined with the expression levels or mutation of known biomarkers,and the patients were divided into 4 subgroups according to their expression level/mutation,and the survival of patients were compared.Result HK2 mRNA was highly expressed in glioma tissues than normal tissues;compared with low grade glioma(LGG),the ex-pression of HK2 mRNA was higher in glioblastoma(GBM).In TCGA and CGGA databases,the expression levels of HK2 mRNA in glioma patients with WHO grade Ⅱ,Ⅲ and Ⅳ were different,with the increase of WHO grade of glioma,the expression levels of HK2 mRNA increased(P<0.05).The expression levels of HK2 mRNA in glioma patients with IDH wild type and IDH mutant type,chromosome 1p/19q co-deletion and chromosome 1p/19q non co-deletion were compared,the differences were statistically significant(P<0.05).Kaplan-Meier survival analysis showed that,in CGGA database,the survival rates of HK2 mRNA high expression group of glioma,LGG,and GBM patients were lower than those in the HK2 mRNA low expression group,and the differences were statistically significant(P<0.05).High HK2 mRNA expression,high NES mRNA expression,no IDH1 mutation and no ATRX mutation indicated poor prognosis.Con-clusion The expression of HK2 mRNA is related to the malignant degree of glioma,and the survival rate of patients with high expression of HK2 mRNA is low.HK2mRNA can be used as a predictor index of prognosis of glioma patients.
