Analysis of the expression of key genes in pancreatic ductal adenocarcinoma based on bioinformatics and their relationship with prognosis
Objective To screen the key genes in pancreatic ductal adenocarcinoma using bioinformatics technology,and analyze their relationship with prognosis.Method Differentially expressed genes(DEG)were screened from pancre-atic ductal adenocarcinoma tissues and adjacent normal tissues of GSE28735 downloaded from the Gene Expression Om-nibus(GEO)database,and the Gene Ontology(GO)functional analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis were performed.The protein-protein interaction(PPI)network was constructed,and key genes were screened by Cytoscape.The mRNA expression of key genes were analyzed,and the relationship between the expression of key genes and the prognosis of pancreatic cancer patients were analyzed.Result A total of 413 DEG were screened,257 up-regulated genes and 156 down-regulated genes.GO analysis showed that DEG were mainly involved in positive regulation of transcription from RNA polymerase Ⅱ promoter,cytosol,protein binding,nucleoplasm and mem-brane.KEGG analysis showed that DEG were mainly enriched in pathways in cancer,salmonella infection,cell cycle,cy-clic adenylic acid(cAMP)signaling pathway,and axon guidance.Five key genes,collagen type Ⅰ alpha 2 chain(COL1A2),periostin(POSTN),fibronectin 1(FN1),collagen type Ⅰ alpha 1 chain(COL1A1),collagen type Ⅲ alpha 1 chain(COL3A1)were identified through the PPI network.All five key genes were highly expressed in pancreatic ductal adenocarcinoma tissues.According to the expression of five key genes,the patients with pancreatic ductal adenocarcino-ma were divided into high expression group and low expression group,the results showed that compared with the low ex-pression group,the overall survival of pancreatic ductal adenocarcinoma patients with high expression of COL1A2,POSTN,FN1,COL1A1 and COL3A1 were significantly shortened,and the prognosis was poor.Conclusion COL1A2,POSTN,FN1,COL1A1 and COL3A1 are all highly expressed in pancreatic ductal adenocarcinoma tissues and are key genes in the occurrence and development of pancreatic ductal adenocarcinoma,and may serve as biological markers for the prognosis of pancreatic ductal adenocarcinoma patients.
