Expression and clinical significance of cyclin-dependent kinase 4 in glioma
Objective To investigate the expression and clinical significance of cyclin-dependent kinase 4(CDK4)in glioma.Method The data in the Genotype-Tissue Expression(GTEx),The Cancer Genome Atlas(TCGA)and Chinese Glioma Genome Atlas(CGGA)databases were mined and analyzed to extract the RNA transcriptome data of CDK4 in gliom.The expression level of CDK4 mRNA in glioma tissues and normal brain tissues was compared,and the relation-ship between the expression level of CDK4 and glioma grade was analyzed.According to the median CDK4 expression levels in TCGA and CGGA databases,patients were divided into CDK4 high expression group and CDK4 low expression group,and the survival of the two groups was compared.Cox regression model was used to analyze the factors affecting the prognosis of glioma patients.The receiver operating characteristic(ROC)curve was plotted to analyze the predictive value of CDK4 for the prognosis of glioma patients,and the nomogram was constructed and verified.According to the median CDK4 expression level in the TCGA database,the patients were divided into CDK4 high expression group and CDK4 low expression group,differential gene analysis was performed for the patients in the two groups,and the Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis were performed on differ-ential genes.Result The expression level of CDK4 mRNA in glioma tissues was significantly higher than that in normal brain tissues(P<0.01).The expression level of CDK4 was positively correlated with glioma grade.The overall survival of CDK4 high expression group was lower than that of CDK4 low expression group.CDK4 was an independent prognostic factor of glioma patients(P<0.01),and had a good predictive effect on prognostic of glioma patients.GO enrichment anal-ysis showed that differential genes were related to mitosis,ion channels,and transmembrane transport,KEGG enrichment analysis showed that differential genes were mainly concentrated in neural active ligand-receptor interactions,calcium sig-naling pathway,cyclic adenylic acid(cAMP)signaling pathway,and cell cycle.Conclusion CDK4 gene plays an impor-tant role in the diagnosis and clinical treatment of glioma.
