摘要
目的 探讨线粒体核糖体蛋白L3(MRPL3)与肺癌细胞增殖及迁移能力的关系.方法 从癌症基因组图谱(TCGA)数据库和在线网站UALCAN获得MRPL3在各肿瘤组织和癌旁组织中的表达水平.构建过表达MRPL3慢病毒转染H1299细胞株,构建敲低MRPL3慢病毒转染HCC827细胞株.采用定量逆转录聚合酶链反应(qRT-PCR)和蛋白质印迹法(Western blot)验证细胞株是否构建成功.采用CCK-8法检测肺癌细胞的增殖能力,Transwell细胞迁移实验检测细胞迁移能力.结果 生物信息学分析显示,MRPL3在肺腺癌组织中的表达水平明显高于邻近正常组织,MRPL3在各肿瘤组织中的表达水平普遍高于癌旁组织.对体外培养肿瘤细胞进行检测,过表达MRPL3肺癌细胞的增殖及迁移能力高于对照组,敲低MRPL3肺癌细胞的增殖及迁移能力低于对照组.结论 MRPL3过表达对肺癌细胞的增殖及迁移起到促进作用.
Abstract
Objective To investigate the relationship between mitochondrial ribosomal protein L3(MRPL3)and pro-liferation,migration of lung cancer cells.Method The expression levels of MRPL3 in tumor tissues and adjacent tissues were obtained from The Cancer Genome Atlas(TCGA)database and UALCAN.The overexpressing MRPL3 lentivirus transfected H1299 cell line was constructed,and the knockdown MRPL3 lentivirus transfected HCC827 cell line was con-structed.Quantitative reverse transcription-polymerase chain reaction(qRT-PCR)and Western blot were used to verify the successful construction of cell lines or not.The proliferative and migration ability of lung cancer cells were detected by CCK-8 assay and Transwell cell migration assay.Result The bioinformatics analysis showed that the expression lev-el of MRPL3 in lung adenocarcinoma tissues was higher than that in adjacent normal tissues,and the expression levels of MRPL3 in various tumor tissues were generally higher than those in adjacent tissues.The proliferation and migration abil-ity of overexpressing MRPL3 lung cancer cells were higher than those of control group,and the proliferation and migra-tion ability of knockdown MRPL3 lung cancer cells were lower than those of control group.Conclusion Overexpression of MRPL3 can promote the proliferation and migration of lung cancer cells.
