Objective To establish and identify the organoid model of high-grade serous ovarian cancer.Method Ovarian cancer tissues of 10 patients with high-grade serous ovarian cancer who underwent ovarian reduction were select-ed.The ovarian cancer tissues were digested into monoplast and cultured in matrix gel for 7 days to construct high-grade serous ovarian cancer organoid model.The morphologic,immunohistochemical characteristics and hematoxylin-eosin(HE)staining results of organoids and primary tumors were compared.Result Organoid models of high-grade serous ovarian cancer were successfully constructed in 10 patients.The optical microscope showed that the organoids were abun-dant and the shapes were like circular 3D stereoscopic structure.HE staining showed that the nuclei of organoids were het-eromorphic at high power lens,and micropapillary structures were formed in local areas.Immunohistochemistry showed strong positive expression of mucin 16(MUC16),strong diffuse positive expression of p53,and immunohistochemical staining of organoids and primary tumors were consistent.Conclusion The successfully constructed high-grade serous ovarian cancer organoid model has obvious epithelial characteristics and is highly similar to the primary tumors,which can be used for drug screening and basic research modeling of ovarian cancer.
