Objective To investigate the SEPTIN9 methylation and E-cadherin expression in advanced gastric cancer and their relationship with prognosis of patients.Method The gastric cancer tissues and adjacent tissues of 52 patients with advanced gastric cancer were collected,the methylation of SEPTIN9 was detected by methylation specific poly-merase chain reaction(MSP)and the relative expression of E-cadherin was detected by Western blotting.The methylation of SEPTIN9 and the relative expression level of E-cadherin in gastric cancer tissues and adjacent tissues,and advanced gastric cancer patients with different clinical characteristics were compared.All patients were followed up for 1 year,the survival of patients during the follow-up period was recorded,the survival of advanced gastric cancer patients with differ-ent methylation of SEPTIN9 and relative expression level of E-cadherin were compared,and the influencing factors for prognosis of patients with advanced gastric cancer were analyzed by Cox risk proportional regression model.Result The positive rate of methylated SEPTIN9 in gastric cancer tissues of advanced gastric cancer patients was significantly higher than that in adjacent tissues,and the relative expression level of E-cadherin was significantly lower than that in ad-jacent tissues,and the differences were statistically significant(P<0.01).The positive rate of methylated SEPTIN9 in gas-tric cancer tissues of advanced gastric cancer patients with moderate and low differentiation and lymph node metastasis were higher than those of patients with high differentiation and no lymph node metastasis,and the differences were statis-tically significant(P<0.05).The relative expression level of E-cadherin in gastric cancer tissues of advanced gastric can-cer patients with moderate and low differentiation and lymph node metastasis were lower than those in patients with high differentiation and no lymph node metastasis,and the differences were statistically significant(P<0.05).The 1-year sur-vival rate of methylated SEPTIN9 positive advanced gastric cancer patients was lower than that of methylated SEPTIN9 negative patients,and the 1-year survival rate of advanced gastric cancer patients with high expression of E-cadherin was higher than that of patients with low expression of E-cadherin,and the differences were statistically significant(P<0.05).Multivariate Cox regression analysis showed that lymph node metastasis,degree of tumor differentiation,methylation of SEPTIN9 and relative expression level of E-cadherin were influencing factors for the prognosis of patients with advanced gastric cancer(P<0.05).Conclusion The positive rate of methylated SEPTIN9 and the relative expression level of E-cad-herin in gastric cancer tissues of patients with advanced gastric cancer are high.SEPTIN9 methylation and E-cadherin ex-pression can reflect the prognosis of patients with advanced gastric cancer,and are related to lymph node metastasis and tumor differentiation.
