Genome-wide Molecular Characterization of G8P[8]-type Group-A Rotavirus in South-central China
Rotaviruses of the G8 genotype have emerged and increased rapidly in China.We undertook genome-wide molecular characterization of seven G8P[8]-type RVA strains collected from children under 5 years of age with acute gastroenteritis in south-central China.Rotavirus-specific amplification combined with high-throughput sequencing was used to sequence the whole genome of 14 G8P[8]-type RVA strains from south-central China.Whole-sequence and genetic variation and evolutionary analyses of the RVA strains were carried out using MEGA11.0,Geneious Prime,Geneious9.0.2 and DNASTAR.Seven G8P[8]-type RVA strains were genotyped as G8-P[8]-I2-R2-C2-M2-A2-N2-T2-E1-H2,which was shown to be a DS-1-like genotype.The phylogenetic tree showed that the seven strains of G8P[8]in south-central China were divided into two major lineages:G8-I(13 strains)and lineage G8-Ⅱ(1 strain).The maximum branching confidence(MCC)tree of VP7 indicated that the average evolutionary rate of G8 was estimated to be 1.4 × 10-3 substitutions/locus/year.In summary,the genotype of G8P[8]and RVA comprised two lineages in China.The average evolutionary rate was estimated to be 1.4 × 10-3 substitutions/location/year.The time of emergence to the epidemic spread of the virus in China will take 1-2 years,and continuous national surveillance of RVA is essential.
