Sequence Analysis of Coxsackievirus A16 in Xianyang,China from 2018 to 2022
This study was aimed to investigate the genetic evolutionary characteristics of coxsackievirus A16(CV-A16)associated hand,foot and mouth disease(HFMD)in Xianyang,China from 2018 to 2022 and to facilitate the establishment of strategies for HFMD control.Surveillance data of HFMD samples in Xianyang,China from 2018 to 2022 were analyzed,then the viral capsid protein 1(VP1)sequences of selected samples were amplified with RT-PCR and sequenced.Full VP1 sequences were determined to create the phylogeny and sequence similarity analysis.During 2018-2022,a total of 402 CV-A16 positive cases were identified,accounting for 28.69%(402/1401)of the laboratory-confirmed cases.VP1-based phylogenetic analysis revealed that 54 sequences belonged to sub-genotype B1,in which 25 belonged to B1a evolutionary branches and 29 belonged to B1b.During 2018-2020 B1b was the major strain,while B1a has become predominant from 2021.Amino acid sequence analysis showed that the B1a contained T164K(25/25),1251V(25/25)and A22T(3/25)mutations,while B1b mainly contained N14S(9/29)and I235V(6/29).CV-A16 was one of the major pathogens of HFMD in Xianyang,China in 2018,2019,2021 and 2022,the dominant strain in Xianyang,China may have converted from B1b to B1a since 2021.The dynamic monitoring of CV-A16 genotypes should be strengthened.
