Rational Design of Vaccines Based on Protein Structure
Thanks to deepening understanding of the structure of virus antigens,structure-based antigen design has become an important means of developing new vaccines.Herein,we summarize the recent developments and research directions of structural vaccinology based on structural biology.We focus on the design method of structural vaccinology based on mutations of amino-acid sites,including the substitution of disulfide bonds,proline substitution,and cavity hydrophobic substitution.We also discuss the specific applications of these methods in the development of vaccines against the human immunodeficiency virus,respiratory syncytial virus,human metapneumovirus,severe acute respiratory syndrome-coronavirus-2,influenza virus,and human papillomavirus.
