高通量测序技术在卡波西肉瘤研究中的应用进展

高通量测序技术,又称二代测序技术,与传统第一代测序技术相比,具有高通量、高分辨率、低成本等优点,已被广泛用于多种疾病和病原微生物学的研究,包括肿瘤及肿瘤相关病毒学领域.高通量测序可通过全面分析复杂的病毒和宿主基因组,探索新的基因亚型及功能,挖掘与病毒发病机制相关的途径,为相关疾病的诊治提供诊疗依据.卡波西肉瘤是一种内皮细胞源性的多灶性肿瘤,卡波西肉瘤相关疱疹病毒(Kaposi's sarcoma-associated herpesvirus,KSHV)为其发病的关键因素,其致病机制目前尚不明确,卡波西肉瘤具有明显地域分布特点,不同地区KSHV具有不同的基因型特征,高通量测序技术目前已被用于KSHV和卡波西肉瘤基因检测以及KSHV分型等研究,该文对其应用及发现进行综述,为卡波西肉瘤发病机制研究及临床诊治提供参考.
Advances in the Application of High-throughput Sequencing Technology in the Study of Kaposi's Sarcoma
High-throughput sequencing technology,also known as second-generation sequencing technology,has the advantages of high-throughput,high-resolution and low-cost compared with the traditional first-generation sequencing technology.It has been widely used in the research of a variety of diseases and pathogenic microbiology,including tumor and tumor-related virology.High-throughput sequencing can comprehensively analyze the complex virus and host genomes,explore new gene subtypes and functions,and the pathways related to the pathogenesis of the virus,so as to provide basis for the diagnosis and treatment of related diseases.Kaposi's sarcoma is a multi-focal tumor derived from endothelial cells.Kaposi's sarcoma-associated herpesvirus(KSHV)is the key factor in the pathogenesis of Kaposi's sarcoma,but its pathogenic mechanism is still unclear.Kaposi's sarcoma has obvious geographical distribution characteristics,and KSHV genotypes in different regions are different.High-throughput sequencing technology has been used for KSHV and Kaposi's sarcoma gene detection and KSHV typing.This article reviews the application and findings of high-throughput sequencing technology,in order to provide reference for the pathogenesis,clinical diagnosis and treatment of Kaposi's sarcoma.
