Study on the Antiviral and Immunomodulatory Effects of Viola Tianshanica Maxim Extract on Mice Infected with Influenza A H1N1 Virus
To clarify the anti-influenza A(H1N1)effect of Viola tianshanica Maxim extract and explore its immunomodulatory mechanism.The mouse pneumonia model induced d by H1N1 infection was divided into normal group,model group,Tamiflu group,and Viola tianshanica Maxim extract 320,160,and 80 mg·kg-1 dose groups.The lung index,viral load in lung tissue,immune factors in lung tissue,immune cell Th1/Th2,Th17/Treg ratio in spleen tissue,total amount and activation of NK cells in spleen tissue were detected;lung pathologic change was observed.Compared with the normal group,the lung index of mice in the model group was significantly increased(P<0.01);the viral load was significantly increased(P<0.01);the contents of TNF-α and TGF-β were significantly increased(P<0.01).,IL-17A and IFN-γ tended to increase but there was no statistical difference.IL-12 basically showed no difference.The ratio of Th1/Th2 increased significantly(P<0.05),and Th17/Treg showed a decreasing trend but no significant difference.The total NK cell amount in the spleen tissue increased significantly(P<0.01),and the activated NK cell content increased significantly(P<0.05).There was a large-scale inflammatory infiltration in the interstitium of the lung tissue,accompanied by obvious stasis.There is bleeding phenomenon,and there is mucus exudation in the interstitium of the lung tissue.Compared with the model group,Viola tianshanica Maxim extract can significantly reduce the lung index of mice(P<0.05);reduce the viral load in mouse lung tissue(P<0.01);reduce the contents of TNF-α and TGF-β,and increase IL-17A,IFN-γ and IL-12 contents(P<0.01).It can increase the number of activated NK cells(P<0.01),and improve lung tissue lesions.Viola tianshanica Maxim extract exerts anti-influenza A(H1N1)virus infection through multiple mechanisms such as reducing lung damage,reducing viral load,inhibiting inflammatory response,regulating key cytokines,and activating immune cells.
