Rapid Detection of SARS-CoV-2 Variants by HRM Technology
To:(ⅰ)establish a method based on high-resolution melting(HRM)curves for the rapid detection of severe acute respiratory-coronavirus-2(SARS-CoV-2)variants;(ⅱ)provide technical support for the rapid detection of SARS-CoV-2 variants.HRM primers were designed for the amino-acid mutation sites G339H/D,D796Y,and V83A of the S gene of SARS-CoV-2.Detection of the nucleic acids of SARS-CoV-2 was undertaken with real-time reverse transcription-quantitative polymerase chain reaction.Thirty-three samples of SARS-CoV-2-positive nucleic acids were detected and compared with the results of next-generation sequencing.The HRM method could be used to distinguish the Delta,Omicron,and XBB variants of SARS-CoV-2.HRM analyses of 33 samples showed that there were two Delta,31 Omicron,and 12 XBB subtypes of SARS-CoV-2.The results of HRM analyses were consistent with those of next-generation sequencing.Our HRM method could be used to accurately detect the Delta,Omicron,and XBB variants of SARS-CoV-2.
