To optimize the extraction process of total flavonoids from tussah's excrement and to inves-tigate the in vitro antioxidant activities,the effects of ethanol volume fraction,extraction tempera-ture,material-liquid ratio,and ultrasonic power on the yield of flavonoids during the extraction process were investigated using the yield of flavonoids as the evaluation index.Based on the results of the one-way test,the Box-behnken central combination design principle was used to optimize the process conditions.The antioxidant capacity of total flavonoids was determined by DPPH and ABTS methods.The optimal extraction process was an ethanol volume fraction of 62%,an extraction tem-perature of 64 ℃,a material-liquid ratio of 1∶23,and an ultrasonic power of 384 w,resulting in a to-tal flavonoid yield of 3.6%.The IC50 of the total flavonoids from the tussah's excrement against DPPH and ABTS free radical scavenging were found to be 0.405 mg/mL and 0.115 mg/mL,respec-tively.The optimized extraction process is stable and reliable,and the total flavonoids obtained from tussah's excrement exhibit good antioxidant activity.
