Optimization of extraction process and in vitro antioxidant activity analysis of total flavonoids from Striga asiatica(L.)O.Ktze
Objective:To optimize the extraction process of total flavones from Striga asiatica(L.)O. Ktze and to analyze its antioxidant activity. Methods:On the basis of single factor test, the ratio of solid to liquid, extraction temperature, ethanol volume fraction, extraction time and extraction times were used as the influencing factors. The extraction rate of total flavones was selected as the evaluation index, and the optimal extraction process was obtained by orthogonal design test and data analysis. In addition, the reducing power of vitamin C to remove DPPH,hydroxyl radical and iron ion was studied. Results:The orthogonal test results showed that the optimal extraction process of total flavonoids was A1B3C3D3,that is,ethanol concentration 55%,solid-liquid ratio 1:50 g/mL,extraction temperature 70℃. Under the optimal conditions, the average extraction rate of total flavonoids was 6 .325%.The total flavones of Striga asiatica(L.)O.Ktze had a certain antioxidant ability, the maximum scavenging rate of DPPH free radical and hydroxyl free radical were 89 .53%and 58 .52%,and the maximum reducing power was 0 .021 . Conclusion:The orthogonal experiment technique is simple and feasible, and can be used to extract total flavonoids from Striga asiatica (L.)O.Ktze. It provides experimental basis for the exploitation and utilization of its resources.
