Determination of dehydrocavir content in different parts of Yanhuanglian medicinal herb by high-performance liquid chromatography
Objective:To establish an efficient liquid chromatography method for determining the content of dehydrocavirin,an effective component of Yanhuanglian medicinal herb;Method:Using a Shiseido Spolar C18 chromatography column(particle size 5)μm.Specification:250mm × 4.6mm,mobile phase:acetonitrile-0.05mol·L-1 sodium dihydrogen phosphate solution=25∶75(6g anhydrous sodium dihydrogen phosphate was prepared with ultrapure water to form a 1L solution),flow rate:1.0mL·min-1,wavelength set at 347nm,column temperature maintained at 30 ℃.Result:The effective ingredient dehydrocavir in the medicinal herb of Yanhuanglian ranges from 0.0241 to 0.3856 μg.There is a good linear relationship within the range of g(r=0.9999,n=5),with an average recovery rate of 99.48%and RSD=1.44%.The content of dehydrocavirin in the roots is higher than that in other parts.Conclusion:This method is fast,accurate,and has strong specificity,providing technical support for the development and quality control of Yanhuanglian medicinal materialsc.
